Expressions of human telomerase mRNA component (hTERC) and telomerase reverse transcriptase (hTERT) mRNA in effusion cytology

Abstract: Telomerase is a ribonucleoprotein enzyme that synthesizes telomeric DNA onto chromosomal ends. Its activity has been reported to be a novel diagnostic marker of malignant potential. However, no study has investigated the diagnostic importance of the expressions of human telomerase mRNA component (hTERC) and telomerase reverse transcriptase (hTERT) mRNA in effusion cytology. We investigated the expressions of hTERC and hTERT using in situ hybridization in 20 malignant cytologic specimens from 20 patients with a… Show more

“…The removed livers were stained with hematoxylin-eosin (HE) for regular histological examination. Also, in situ hybridization of TNF mRNA was performed as described previously [16] . Detailed information is described in the online supplement.…”

“…The diagnostic value of the ELISA‐based TRAP assay may be undermined in tuberculosis endemic areas, and telomerase activity should be interpreted particularly cautiously wherever the annual risk of tuberculous infection is still high, for example, in Taiwan 29 . Recently, the in situ TRAP assay and in situ hybridization for human telomerase mRNA component and telomerase reverse transcriptase mRNA have been described 30,31 . These advanced techniques may be helpful for distinguishing between inflammatory and malignant cells in a non‐malignant pleural effusion with detectable telomerase activity.…”

“…The sense probe was used as a negative control. To confirm the presence of intact RNA, replicate slides from each sample used for TNF ‐ alpha expression were also evaluated for the expression of housekeeping genes using biotinylated RNA‐positive control oligonucleotides as described previously . The hybridization was performed overnight at 37°C.…”
