Extended wavelength anisotropy resolved multidimensional emission spectroscopy (ARMES) measurements: better filters, validation standards, and Rayleigh scatter removal methods

Abstract: Anisotropy resolved multidimensional emission spectroscopy (ARMES) provides valuable insights into multi-fluorophore proteins (Groza et al 2015 Anal. Chim. Acta 886 133-42). Fluorescence anisotropy adds to the multidimensional fluorescence dataset information about the physical size of the fluorophores and/or the rigidity of the surrounding micro-environment. The first ARMES studies used standard thin film polarizers (TFP) that had negligible transmission between 250 and 290 nm, preventing accurate measurement… Show more

“…Irrespective of the scanning mode, an EEM is obtained. More recently, TSFS/EEM has been combined with anisotropy measurements to generate a 4-dimensional data matrix [7,9,10]. If generated via TSFS, the 4-dimensional data matrix comprises excitation wavelength (λ ex ), wavelength offset (Δλ), Intensity (I ex/Δλ ), and anisotropy (r ex/Δλ ).…”

“…This offers an attractive, mathematical alternative to the more laborious standard addition or modified standard addition methods [100]. Finally, PARAFAC has also been used to model Rayleigh scatter in EEM data, which can then be used to remove the scatter signal [10,101].…”

“…Furthermore, Trp emission is very sensitive to the local physicochemical environment, and emission changes can be used to monitor conformational change, denaturation, and binding (Ghisaidoobe & Chung, 2014). To extract more information from in situ measurements, one can use multidimensional measurements like excitation-emission matrix (EEM) and anisotropy resolved multidimensional emission spectroscopy (Casamayou-Boucau & Ryder, 2017;Groza, 2016;Groza, Li, & Ryder, 2015). Here, we first investigate the use of polarized excitation-emission matrix (pEEM) spectroscopy to measure protein structural changes and other physicochemical changes caused by PEGylation.…”

“…The ARMES measurement wavelength range can be extended using polarizers with better UV transmittance and enable the acquisition of data in the important tyrosine/tryptophan region [36]. Polarizer efficacy was confirmed using a perylene standard which has two different emitting states with different fundamental anisotropies of 0.4 (S1) and -0.2 (S2) [37,38], and the measured anisotropy was significant in viscous glycerol solution: 0.197 and -0.031 ± 0.002 at 25°C [39].…”
