Extracellular ATP mediates the late phase of neutrophil recruitment to the lung in murine models of acute lung injury

Abstract: Acute lung injury (ALI) is a severe inflammatory condition whose pathogenesis is irrevocably linked to neutrophil emigration to the lung. Activation and recruitment of neutrophils to the lung is mostly attributable to local production of the chemokines. However, much of our understanding of neutrophil recruitment to the lung is based on studies focusing on early time points after initiation of injury. In this study, we sought to evaluate the extended temporal relationship between neutrophil chemotactic factor … Show more

“…Consistent with this hypothesis, we observed decreased AMPactivated protein kinase (AMPK) activation and increased lipid synthesis in EtOHexposed lungs and in cultured AEII cells. Bronchoalveolar lavage (BAL), total cell counts, and differential cell counts were performed as previously described (21).…”

“…Murine Model of Acute Lung Injury (ALI)-ALI was induced by instilling 100 l lipopolysaccride (LPS, 100 mcg) or 50 l of HCl (0.1N) into posterior oropharyngeal space of anesthesized mice as previously described (21,22). Control mice received it.…”

“…Total cell count was determined with a TC20 automated cell counter (Bio-Rad Laboratories) while differential counts were performed on cells cyto-centrifuged onto glass slides (Fisher Scientific). Total protein concentration in the BAL fluid was determined using the Pierce TM BCA assay kit (Thermo Scientic, Rockford, IL) as previously described (21).…”
