Extracellular Phospholipase A2 and Histamine Release from Rat Peritoneal Mast Cells

Abstract: Phospholipase A2 (PLA2) from cobra (Naja naja) venom and PLA2 from porcine pancreas accelerated IgE antibody-mediated histamine release from rat peritoneal mast cells. These enhancements were clearly abrogated by heating the enzymes and pretreatment with parabromophenacyl bromide, mepacrine and anti-flammin. Indomethacin (cyclooxygenase inhibitor) and AA-861 (lipoxygenase inhibitor) did not affect the enhancement by PLA2. These results indicate that extracellular PLA… Show more

“…Preincubation of pancreatic PLA 2 with AF‐1blocks its activity on mast‐cell degranulation. 65 AFs interact with lipid‐water interfaces 66 and inhibit the increase in fluorescence of Trp 3 in porcine pancreatic sPLA 2 that is normally triggered by binding to micellar substrates. 25 All in all, the most likely explanation for these observations is that, like UG, AFs also inhibit either the interfacial activation or the interaction of sPLA 2 's with lipid‐water interfaces.…”
