Extracellular vesicles for liquid biopsy in prostate cancer: where are we and where are we headed?

Abstract: Background:Extracellular vesicles (EVs) are a heterogeneous class of lipid bound particles shed by any cell in the body in physiological and pathological conditions. EVs play critical functions in intercellular communication. EVs can actively travel in intercellular matrices and eventually reach the circulation. They can also be released directly in biological fluids where they appear to be stable. Because the molecular content of EVs reflects the composition of the cell of origin, they have recently emerged a… Show more

“…G. Zhang & Grizzle, 2014; Zhao et al, 2017). Exosomes secreted by tumor cells can be found in the blood and urine, which makes analysis of exosomal content attractive for early cancer diagnosis in “liquid biopsy” (Kai et al, 2017; Minciacchi, Freeman, & Di Vizio, 2015; Minciacchi, Zijlstra, Rubin, & Di Vizio, 2017; A. Sharma, Khatun, & Shiras, 2016; Yokoi, Yoshioka, & Ochiya, 2015; Yoshioka et al, 2014; W.…”

“…Recent literature has established a critical role for these nano‐sized vesicles in intercellular communication, tumor growth, and progression . Importantly, exosomes are also emerging as important tools for disease diagnosis and prognosis . McKiernan et al developed a urine exosome‐based assay for better prognosis of the disease .…”

“…Despite these limitations, molecular markers such as mRNA, proteins, lipids and small RNAs remain a promising strategy for diagnosis and subtyping of cancer or other chronic diseases. With the advancements in the field of high-throughput data and the increased attention to the issues of data normalization and statistical modelling, some drawbacks of biomarker mining may be overcome in a foreseeable future (Minciacchi, et al, 2017).…”
