Extraintestinal pathogenic Escherichia coli utilizes the surface-expressed elongation factor Tu to bind and acquire iron from holo-transferrin

Abstract: Extraintestinal pathogenic Escherichia coli (ExPEC) is a common anthropozoonotic pathogen that causes systemic infections. To establish infection, ExPEC must utilize essential nutrients including iron from the host. Transferrin is an important iron source for multiple bacteria. However, the mechanism by which ExPEC utilizes transferrin remains unclear. In this study, we found that iron-saturated holo-transferrin rather than iron-free apo-transferrin promoted the vitality of ExPEC in heat… Show more

“…Reports have shown that immunization with NanA, Tuf, and Ply induced protective effects against focal and lethal infections with different S. pneumoniae serotypes, which have been targeted by various scientists to develop a potential vaccine ( 48 50 ). Bioinformatic analysis, as one of important and prominent computational tools, could offer deep mechanistic insights into the complexity of biological systems and obtain high-performance candidate vaccines.…”

“…The co-existence of the other virulence-related genes was found in the V. cholerae genomes as well (Table S9). For example, the V. cholerae 7-6-5 genome encoded the cell surface-expressed elongation factor EF-Tu (Vc 7_6_5_2186, Vc 7_6_5_2235, Vc 7_6_5_3026, and Vc 7_6_5_3364) [44], and lipid mediator receptors connected lipo-oligosaccharides (LOSs) (Vc 7_6_5_0960) [45]. The LOSs of Histophilus somni may act as an adhesin and an endotoxin that signals through toll-like receptor 4 and NF-κB to cause inflammation in the host [45].…”

“…Homologous proteins sequences were uploaded to a CLUSTAL Omega program available through UniProt to generate alignments for annotation (Sievers et al, 2011). Additional known and putative binding substrates for each protein were found from literature review (Sun et al, 2022, Harvey et al, 2019, Tomoyasu et al, 2012, Candela et al, 2010.…”
