Eye drop delivery of nano‐polymeric micelle formulated genes with cornea‐specific promoters

Abstract: Cornea epithelium- and stroma-specific gene expression could be achieved using cornea-specific promoters of keratin 12 and keratocan genes, and the gene was delivered with PM formulation through non-invasive, eye drop in mice and rabbits. The transfection mechanism of plasmid-PM may involve endocytosis and particle size dependent paracellular transport.

“…Following topical drop administration, the gene-encapsulated polymeric micellar system improved the mRNA levels of bcl-x(L) by 2.2-fold and reduced corneal apoptosis. 115 From these studies, it is evident that an aqueous nanomicellar formulation of drug or genes can improve permeability in anterior tissues.…”

“…The core-shell structure may notably enhance the apparent aqueous solubility, leading to a greater concentration gradient favourable for diffusion, and also provide sustained release since the dilution factor in the lachrymal fluid is less than in other administration routes [16]. Polymeric micelles have been shown to be suitable ocular carriers for anti-inflammatory drugs [17] or even plasmids and genes [18,19]. Transcorneal permeation studies through excised rabbit cornea indicated that non-steroidal anti-inflammatory drug formulations in polymeric micelles can enhance about twofold drug permeation compared with that of an aqueous suspension of the same concentration because the dissolution step is overcome [17].…”

“…Os polímeros biodegradáveis apresentam vantagens sobre os não-biodegradáveis, pois não necessitam ser removidos cirurgicamente após o término de sua ação [2] . A versatilidade proporcionada pelos polímeros sintéticos permite obter micropartículas com perfis de liberação específicos possibilitando administrar por diversas vias de administração [10,11] , como parenteral [15] , oral [16] ou oftálmica [17] .…”
