Fabrication of Silica-Coated Gold Nanorods Functionalized with DNA for Enhanced Surface Plasmon Resonance Imaging Biosensing Applications

Abstract: A novel method for preparing gold nanorods that are first coated with a thin silica film and then functionalized with single stranded DNA (ssDNA) is presented. Coating the nanorods with 3-5 nm of silica improves their solubility and stability. Amine-modified ssDNA is attached to the silicacoated gold nanorods via a reductive amination reaction with an aldehyde trimethoxysilane monolayer. The nanorods exhibit an intense absorption band at 780 nm, and are used to enhance the sensitivity of surface plasmon resona… Show more

“…Gold nanorods (GNRs) are nanoscale materials that possess interesting optical properties and unique application potentials particularly in photothermal therapy, [1,2] biosensing, [3] molecular imaging, [4][5][6] and in gene delivery [7] for cancer therapy. These various potential applications require that the nanorods be functionalized to attach different biochemical groups.…”

“…[13,14] Various strategies have been developed to functionalize gold nanorods with a variety of different ligands to reduce their cytotoxicity. [12,[15][16][17][18][19][20] Further reports have dealt with the problems of stability and biocompatibility of Au NRs by using various polymer shells, [21] polyelectrolytes, [22,23] peptides, [24] surfactants, [25] and lipids [14,26] to modify the Au NR surface. Partial replacement of CTAB was qualitatively confirmed in many of these cases, but the exact surface composition of nanorods and the amount of residual CTAB was either unknown or not possible to determine.…”

“…[59][60][61] For metallic nanoparticles such as gold nanorods that can support photothermal effect, the heat converted from absorbed light can be applied actively to release drugs. Hence gold nanorods have been employed as drug delivery vehicles in vitro, 62) although they have been rarely assessed for in vivo delivery.…”
