Three new series of 4-substituted-5-alkylidene-2,5-dihydrofuran-2-ones were synthesized. The in vitro activity test results showed that some of them exhibited good antibacterial and cytotoxic activities. Among them compound5cshowed the most potent antibacterial activity againstEscherichia coliwith the MIC value of 20.00 μg/mL. Compound9cshowed good cytotoxic activity against Ec9706 cells withIC50value of 19.39 μM, better than that of the reference compound fluorouracil (IC50=37.74 μM).

A new method of preparation for natural product Uncinine was reported. According to the method, a series of novel butenolides derivatives were designed and synthesized based on the natural product Uncinine. Most of the synthetic compounds showed significant anti proliferation activity against MGC-803. Among of them, (Z)-5-(4-(tert-butyl)benzylidene)-3-(morpholinomethyl)furan-2(5H)-one (9l) showed potent anticancer effect with IC 50 of 2.9 μmol/L in gastric cancer cell MGC803 and showed good selectivity to gastric cancer cells MGC803, HGC27, SGC7901 and less cytotoxicity in normal gastric epithelial cell line (GES1). The research on the molecular level suggests that the mechanism of compound 9l inducing apoptosis in gastric cancer cells is partially dependent on activation of caspase-9/3.

Over the past decades, an increasing number of these compounds have been isolated from various natural sources. Members of this class of substances vary greatly in structural complexity and functionality. Besides that, many of them have been shown to display a wide range of biological activities. The g-alkylidenebutenolides has attracted the attention of the organic synthetic chemists resulting in the development of a variety of process to synthesize them. This article review is concerned with the different approaches that can be utilized to prepare g-alkylidenebutenolides.Keywords: g-alkylidenebutenolides; α,β-unsaturated lactones; butenolides. INTRODUÇÃOButenolídeos correspondem a g-lactonas insaturadas, podendo apresentar anéis dos tipos I ou II (Figura 1).Certos D 3-butenolídeos que possuem um grupo alquilideno localizado na posição g (estrutura geral III, Figura 1) são conhecidos genericamente como g-alquilidenobutenolídeos.Durante as últimas décadas, um grande número de g-alquilidenobutenolídeos foi isolado de várias fontes naturais. Muitos membros dessa família, alguns deles mostrados na Figura 2, apresentam importantes atividades biológicas, incluindo atividade antibiótica, apresentada pelos compostos protoanemonina (1), 1 lissoclinolídeo (8) 6 e rubrolídeos (15-20); 11 inibição da biossíntese do colesterol, observada com as substâncias xerulina (5), ácido xerulínico (6) e diidroxerulina (7); 5 citotoxicidade, observada com os goniobutenolídeos A (10) e B (11) 8 e os nostoclídeos I (12) e II (13). 9 Outros exemplos dessa classe de butenolídeos são os compostos 29-31. A substância 29 é conhecida por ativar a germinação de sementes, 13 enquanto a pirroxantina (30) 14 e peridinina (31) 15 correspondem aos g-alquilidenobutenolídeos de maior complexidade estrutural já isolados e desempenham papel importante na fotossíntese de algas e dinoflagelados, respectivamente.Os g-alquilidenobutenolídeos têm sido considerados atrativos alvos sintéticos devido à diversidade estrutural que apresentam, bem como às suas propriedades biológicas. Como consequência, vários procedimentos sintéticos vêm sendo desenvolvidos para o preparo dessas substâncias. 16 Nesta revisão são descritos vários métodos que podem ser utilizados para a síntese de g-alquilidenobutenolídeos.
