Facilitatory and Inhibitory Effects of Harmaline on the Tryptophan-Induced 5-Hydroxytryptamine Syndrome and Body Temperature Changes in Pargyline-Pretreated Rats

Abstract: ABSTRACT-The effects of harmaline on tryptophan-induced 5-hydroxytryptamine (5-HT) syndrome and body temperature changes in pargyline-pretreated rats were investigated. When administered i.p. 60 min after pargyline treatment (50 mg/kg, i.p.), tryptophan, at 100 mg/kg but not 10 mg/kg, induced the 5-HT syndrome. Tryptophan at 100 mg/kg also produced hypothermia followed by hyperthermia in pargylinepretreated rats. Administration of harmaline (10 mg/kg, i.p.) 30 min after pargyline not only potentiated the 100 m… Show more

“…For instance, citalopram, a selective serotonin reuptake inhibitor, has been reported to augment harmaline-induced tremor 35 . Additionally, after harmaline injection, elevated level of 5-HT level was observed in striatum, cortex, hypothalamus, hippocampus 36,37 and brainstem 35 . Furthermore, sertraline escitalopram, one of the SSRI, was implicated in inducing movement disorders, including dystonia, akathisia, parkinsonian symptoms in a 38-year-old male patient 38 .…”

“…11 Harmala alkaloids exhibit reversible and selective inhibition of MAO-A, which differentiates them from irreversible pharmaceutical MAOIs. [18][19][20] Reversible MAOIs, such as moclobemide, have a reduced propensity to lead to serotonin syndrome and hypertensive crises. [21][22][23] Harmine has been shown to produce serotonin syndrome and myoclonic syndrome in rodents injected with tryptophan.…”

“…Also, the basic mechanisms of action of these agents on particular molecular targets are poorly understood, or they are simply not directed to a particular target. β‐Carboline alkaloids are such a large group of natural and synthetic indole alkaloids, of great pharmacological significance, including in cancer and tumor treatments; hence, numerous studies have been focused on their biological effects …”
