Several studies have shown a role for the involvement of interleukin (IL)-1 gene cluster polymorphisms in the risk of periodontal diseases. In the present study, we tested polymorphisms, derived from genes of the IL-1 cluster, for association with generalized aggressive periodontitis (GAP) through both allelic association and by constructing a linkage disequilibrium (LD) map of the 2q13-14 disease candidate region. The IL-1RN (VNTR) genotype distribution observed was significantly different in GAP and control subjects (P ¼ 0.019). We also observed some evidence for an association between GAP and the IL-1B þ 3953 polymorphism (P ¼ 0.039). The pattern of association in the region, represented as an LD map, identifies a recombination hot area between the IL-1B þ 3953 and IL-1B À511 polymorphisms. Multilocus modelling of association with disease gives a location for the peak association at the IL-1B þ 3953 marker, although support for the peak is not significant. Haplotype analysis identifies a IL-1B þ 3953 -IL-1B À511 haplotype as having the lowest P-value in the region. Recognition of the presence of a recombination hot area between the IL-1B þ 3953 and IL-1B À511 polymorphisms will have an important bearing on future efforts to develop higher resolution SNP analysis in this region for both this and other diseases for which this cluster is implicated.

Several studies have investigated the genetic polymorphisms for cytokines as potential genetic markers for periodontitis. The aim of this study was to determine the prevalence of IL1 cluster genes polymorphisms and their association with chronic periodontitis in the Macedonian population. The group of 114 unrelated Macedonian subjects with chronic periodontitis and 301 periodontitis-free Macedonian subjects were studied. DNA was isolated from peripheral blood leukocytes by phenol-chloroform extraction method. Cytokine genotyping was performed by PCR-SSP. The population genetics analysis package (PyPop) was used for analysis of the cytokine data for this report. Crude odds ratio (OR) was calculated as estimates of the relative risk with 95 % confi dence interval (CI). Genotype frequency of IL1B -511/C:T was signifi cantly higher in patients with periodontitis than in controls (OR=2.11, 95 % CI=1.35-3.32, p=0.001). IL1 cluster gene haplotype frequencies of TTTCT and TCTTT were associated with higher risk for periodontitis (OR=5.06, 95 % CI=1. p<0.0014 and OR=8.35, p<0.002, respectively). No signifi cant association of IL1 composite genotype (IL1 -889A:IL1B +3962) with periodontitis in Macedonians was found. The latter association was found to be signifi cant in genotype IL1B -511/C:T, haplotype TTTCT, and haplotype TCTTT, but without signifi cant association in IL1 composite genotype (Tab. 5, Ref. 43). Genetik polimorfizmler ile çeşitli medikal hastalıkların, kronik immün ve enflamatuvar durumların ilişkilerini inceleyen çalışmalar son on yılda bir artış göstermiştir. Günümüzde periodontal araştırmalar bu büyüyen alana katkıda bulunmaktadırlar. Bu yeni araştırmalar, gen ürünlerinin birbirleri ile olan fonksiyonel ilişkileri ve çevresel ajanlarla olan ilişkilerini anlamaya katkı sağlayan, genomu anlama yönünde de etkilemektedir. Bu araştırmaların sonuçları ışığında, periodontitisi de içine alan birçok hastalık için genetik temellerinin olduğuna dair kanıtlar ortaya çıkmaktadır. Bu kanıtlar da, hastalıkların genetik temellerini anlarsak hastalık riskini belirlemeye yönelik genetik testler ve etiyoloji temelli tedavilerin gelişimi de yakın zaman da gerçekleşecektir, fikrini desteklemektedir. Sonuç olarak, periodontal hastalıkların hastalık riskini belirlemede kullanılan genlerin allelik çeşitliliğini tanımlamak oldukça önemlidir.

Abstract: The lack of any association between the IL1 polymorphisms and GEOP, in the population presented here, brings into doubt the usefulness of these candidate genes as markers of susceptibility to this form of periodontitis.
