Fast Analytical Methodology Based on Mass Spectrometry for the Determination of Volatile Biomarkers in Saliva

Abstract: We report a methodology for the rapid determination of biomarkers in saliva. The method is based on direct coupling of a headspace sampler with a mass spectrometer. The saliva samples are subjected to the headspace generation process, and the volatiles generated are introduced directly into the mass spectrometer, thereby obtaining a fingerprint of the sample analyzed. The main advantage of the proposed methodology is that no prior chromatographic separation and no sample manipulation is required. The following… Show more

“… 34 2-Ethyl-1-hexanol was also found exclusively in lung cancer patients’ saliva samples but not in other cancer patients’ saliva. 35 It was postulated that alcohols are produced by the oxidation of alkanes by cytochrome p450 (CYP450), a group of enzymes that are over-activated in lung cancer tissue. 32 As alkanes result from lipid peroxidation resulting from oxidative stress, the increased level of these two alcohols could be the result of increased oxidative stress and upregulated CYP450.…”

“…Indeed, salivary proteomics is an active research field where a variety of analytical techniques are being developed and applied to profile the proteomes of saliva samples. 5, 11-14 In contrast, there are only a few reports focusing on metabolome analyses of saliva using NMR 15-19 , gas chromatography mass spectrometry (GC-MS) 3, 20, 21 , liquid chromatography MS (LC-MS) 3, 22-25 , and capillary electrophoresis MS (CE-MS). 26 The major challenge in saliva metabolome profiling lies in the relatively low abundance of metabolites present in saliva.…”

“…Urinary mercapturic and mandelic acid metabolites will be included in future NHANES assessments (Alwis et al 2012). A few studies have measured styrene in human saliva and breast milk (Blount et al 2010; Sanchez et al 2012). …”
