Fast and improved separation of major coumarins inAmmi visnaga(L.) Lam. by supercritical fluid chromatography

Abstract: The first supercritical fluid chromatography method for the determination of five major coumarins (dihydrosamidin, visnadin, samidin, khellin, and visnagin) in Ammi visnaga fruits is described. Their baseline separation was possible in less than 5 min by using a UPC HSS C SB column with 1.8 μm particle size and a mobile phase comprising CO , methanol, acetonitrile, and diethylamine. The type of stationary phase used was of particular relevance because, except for the selected one, the others did not resolve th… Show more

“…The frequent occurrence and biological activity render phenolic compounds to one of the most interesting ingredients in plant kingdom. In the last few years an increasing number of analytical applications were investigated focusing on flavonoids [51,52], coumarins [53][54][55], kavalactones [56], anthraquinones [57], and cannabinoids [58]. Huang et al [51] presented very recently a first SFC-DAD-UV method for the analysis of 12 flavonoids, among them flavones, flavanols, as well as mono-und diglycosides.…”

“…A few SFC methods have been published up to now for the determination of coumarins. Pfeifer et al [53] presented a validated SFC-DAD-UV method for the determination of eight congeners in Angelica dahurica roots, Kim et al [54] resolved two pyranocumarins in Angelica gigas roots, and Winderl et al [55] succeeded in the first complete separation of all coumarins in Ammi visnaga fruit.…”

“…On the other hand, thanks to its faster analysis time and low solvent consumption, the SFC technique is a greener technique and an alternative to conventional HPLC [ 23 ]. Moreover, SFC is proving to be a valid alternative approach to investigating coumarins, furocoumarins and polymethoxyflavones [ 24 , 25 , 26 , 27 , 28 , 29 , 30 ]. SFC was already successfully adopted for the analysis of eight furocoumarins in four cosmetics after sample extraction [ 28 ].…”

“…Although a number of analytical studies using UPC 2 (with or without hyphenation to MS) regarding the quantitation of pharmaceutically relevant natural product structure classes have been published [19,[27][28][29][30][31], hops constituents have not been addressed by SFx techniques so far. Hence, the aim of the present study was to establish the first SFx-based protocol for the identification and quantitation of the five characteristic prenylated compounds, i.e., hulupinic acid (1), 8-prenylnaringenin (2), 6-prenylnaringenin (3), isoxanthohumol (4), and xanthohumol (5) (▶ Fig.…”
