Fast Lymphoid Reconstitution After Vascularized Bone Marrow Transplantation in Lethally Irradiated Rats1

Abstract: The results of our studies indicate that the presence of stromal cells in their close relationship with stem cells is essential for the fast lyphohematopoietic repopulation of irradiated recipients. The population of CD4+dull cells may represent immature cells. These cells were not found in MLN of VBMT rats. All MLN CD4+ cells represented the "bright" subset, what suggests that the process of cell maturation may occur in the lymphoid organs.

“…85 The vascularized BM transplant (VBMT) model has been recognized as a better source for hematopoetic cell reconstitution than transplantation of cellular BMCs. 86 In order to achieve successful BMC transplantation, irradiation or other myelosuppressive therapy was used to make space for donor BM engraftment and maintenance; in addition, subsequent immunosuppressive agents were used to prevent the rejection of allogenic cells. 87 The uniqueness of VBMT stems from the fact that it provides its own intact space along with its own microenvironment of supportive stromal cells.…”

“…Their nonmyeloablative therapy model was combined with ALS, tacrolismus, low-dose TBI, and BMT. 86 They used ALS and a limited course of tacrolismus in order to decrease the amount of radiation necessary to achieve marrow engraftment and to limit GVHD. 91,92 Cyclosporine-A prevented immediate donor limb rejection while ALS created the space needed for donor bone engraftment.…”

“…Therefore, by providing the stromal hematopoietic microenvironment as well as pluripotent progenitor cell source, the VBMT seems to be more efficient and essential for the post-transplant lymphopoiesis when compared with cellular BM transplant. 20,[75][76][77] Finally, VBMT has been proved to prolong the survival of skin graft from the same donor. 78 Whether the donor BM in a human hand transplant might induce mixed chimerism in the recipient is unknown.…”
