Fc receptor-like 3 (−169T>C) polymorphism increases the risk of tendinopathy in volleyball athletes: a case control study

Abstract: BackgroundTendinopathy pathogenesis is associated with inflammation. Regulatory T (Treg) cells contribute to early tissue repair through an anti-inflammatory action, with the forkhead box P3 (FOXP3) transcription factor being essential for Treg function, and the FC-receptor-like 3 (FCRL3) possibly negatively regulating Treg function. FCRL3 –169T>C and FOXP3 –2383C>T polymorphisms are located near elements that regulate respective genes expression, thus it was deemed relevant to evaluate these polymorphisms as … Show more

“…84 Furthermore, Treg are also involved in the inflammatory status in both OA and tendinopathy. 85,86 Herein, six of the identified miRNAs were found to be able to modulate Treg polarization and stabilization. miR-146a-5p contrasts the polarization toward inflammatory Th1, 87 miR-99a-5p affects inducible Treg polarization by targeting mammalian target of rapamycin (mTOR), 88 and miR-10a-5p also targets Treg by stabilizing the expression of FoxP3.…”

“…8 studies investigating genes variations associated with rotator cuff tears were included in this review. 6 studies were focused on candidate genes [1924] and 2 studies were GWASs [25, 26].…”

“…As demonstrated in in-vivo studies, the immune cells play a key role in the pathophysiology of rotator cuff tears. Salles et al found an increased risk of tendinopathy associated with Fc receptor-like 3 polymorphism ( FCRL3 − 169 T > C ) [24]. FCRL3 is a glycoprotein of the immunoglobulin receptor superfamily, expressed in Treg cells that may play a role as a negative regulator of Treg function [4042].…”

“…Single nucleotide polymorphisms (SNPs), a variation of the nucleotide at a single position in DNA sequence, involved with inflammatory process were associated as non-modifiable factors for developing tendinopathies [ 6 8 ]. Pro-inflammatory cytokines, such as tumor necrosis factor alpha (TNF-α), and growth factors have been implicated as a mechanism in early stages of tendinopathies [ 1 , 9 ].…”
