Immature zygotic embryos of the sunflower inbred line 'Ha 300', cultivated on a modified MS medium containing 6-benzyladenine and a high amount of sucrose, regenerated fertile plants via direct somatic embryogenesis. Plant regeneration from immature sunflower embryos is generally characterized by a relatively high experimental variability resulting from the interactions of multiple factors. We present here a study of some of the factors acting on the donor plants and their influence on the capacity to regenerate plants.Repeated experiments during a 2-year period with greenhouse-grown as well as field-grown plants led to the following conclusions: (i) The use of pesticides, unavoidable in the greenhouse, is compatible with routine regeneration of fertile plants, (ii) The plant growth retardants tested were useful for the production of healthy plants in the greenhouse and had no effect on the regeneration capacity.

At the beginning of the new millennium, sunflower relied on traditional non-GM varieties, which could be a possible disadvantage in relation to other oilseed crops. Different transformation protocols allowed obtaining desirable transgenic traits: the oxalate-oxidase expressing gene for fungal disease control, glyphosate tolerance by expressing Agrobacterium gene cp4, and the Bt toxin gene cry1 to control Lepidoptera. Ammonium absorption, other herbicide tolerance, Cry1 variants and the CpT1 gene for pest control are new targets. Quality can be improved through a modified protein and latex biosynthesis. The high cost of the GM crop approval process, increased by the probability of gene flow with wild relatives, is a constraint for the transgenic sunflower release. Strategies designed to minimize environmental risks and changes in consumer perception are needed before sunflower transgenic varieties become acceptable.

En esta investigación se desarrolló una técnica in vitro para producir y propagar híbridos intergenéricos a partir de semillas inmaduras producto del cruzamiento del girasol cultivado Helianthus annuus L. con la especie silvestre Tithonia rotundifolia (Mill.) S. F. Blake. Para el rescate de embriones inmaduros en cultivo de tejidos el mayor éxito se obtuvo con el uso secuencial de tres medios basados en el de Murashige-Skoog: a) con adición de aminoácidos y el regulador de crecimiento BAP para germinación, b) sin aminoácidos y con BAP para micropropagación, y c) sin vitaminas y con el regulador de crecimiento ANA para enraizamiento. Los aminoácidos se agregaron como fuente de nitrógeno orgánico. A partir de una plántula se lograron obtener 130 en buenas condiciones de sanidad. Veinte de ellas se llevaron a aclimatación, y solamente una generó botones y llegó a floración inconclusa, por lo que aún se requiere estandarizar las condiciones de aclimatación. En particular, se necesita establecer tratamientos que permitan definir con claridad las condiciones específicas para aclimatar el híbrido, a través de la prueba de diferentes mezclas de sustratos y soluciones nutritivas bajo condiciones ambientales controladas. Se recomienda probar el manejo de retardantes y aminoácidos, así como realizar modificaciones en la intensidad de luz.
