FGF Family: From Drug Development to Clinical Application

Abstract: Fibroblast growth factor (FGF) belongs to a large family of growth factors. FGFs use paracrine or endocrine signaling to mediate a myriad of biological and pathophysiological process, including angiogenesis, wound healing, embryonic development, and metabolism regulation. FGF drugs for the treatment of burn and ulcer wounds are now available. The recent discovery of the crucial roles of the endocrine-acting FGF19 subfamily in maintaining homeostasis of bile acid, glucose, and phosphate further extended the act… Show more

“…All FGFs, except FGF11, FGF12, FGF13, and FGF14 act through their specific receptors (FGFRs), including mitogenesis and non-mitogenic effects (Yun et al, 2010). The FGF ligands elicit their activity by using four transmembrane tyrosine kinase receptors FGFR1, FGFR2, FGFR3, and FGFR4 (Hui et al, 2018). The activated FGFRs will transduce the signals through three dominant pathways including RAS/MAPK, PI3k/AKT, and PLCγ (Yun et al, 2010;Li et al, 2016).…”

“…The protein encoded by this gene is a member of the fibroblast growth factor (FGF) family. FGF family members bind heparin and possess broad mitogenic and angiogenic activities [22] . FGF2 has also been implicated in diverse biological processes, such as limb and nervous system development, wound healing, and tumor growth [23] .…”

“…The preliminary mechanisms through which aFGF promotes the wound healing include aFGF-stimulated secretion of TGF-b and PCNA proteins from the damaged skin and the promotion of capillary and fibroblast proliferation (Xie et al, 2011). Our group previously developed a lyophilized rh-aFGF, which was approved by the Chinese Food and Drug Administration in 2006 for the treatment of deep, partial-thickness burns and skin graft donor sites (Ma et al, 2007;Hui et al, 2018a). This marketed rh-aFGF is a topical lyophilization agent, applied in the form of a spray.…”
