FgMon1, a guanine nucleotide exchange factor of FgRab7, is important for vacuole fusion, autophagy and plant infection in Fusarium graminearum

Abstract: The Ccz1-Mon1 protein complex, the guanine nucleotide exchange factor (GEF) of the late endosomal Rab7 homolog Ypt7, is required for the late step of multiple vacuole delivery pathways, such as cytoplasm-to-vacuole targeting (Cvt) pathway and autophagy processes. Here, we identified and characterized the yeast Mon1 homolog in Fusarium graminearum, named FgMon1. FgMON1 encodes a trafficking protein and is well conserved in filamentous fungi. Targeted gene deletion showed that the ∆Fgmon1 mutant was defective in… Show more

“…Mutation of the endocytic pathway related components often showed defects in vacuole fusion and vesicle trafficking in different phytopathogens. These include the Rab GTPase FgRab7, the guanine nucleotide exchange factor FgMon1, the Vesicle trafficking in F. graminearum 1443 HOPS complex subunit FgVps39, the retromer complex subunit FgVps35 and the SNARE protein FgVam7 in F. graminearum (Li et al, 2015;Zheng et al, 2016;Li et al, 2017a), and the SNARE proteins MoVam7 and MoSec22, and MoVps41 in the rice blast fungus (Song et al, 2010;Dou et al, 2011;Zhang et al, 2017), the SNARE protein UmYup1 in Ustilago maydis (Fuchs et al, 2006). All of these gene deletion mutants showed fragmented vacuoles and was defective in fungal development and infection.…”

“…In our previous study, Rab GTPases play an essential role in the development of head blight of wheat and mycotoxin production in F. graminearum (Zheng et al, ). A recent study shows that FgMon1, a FgRab7 GEF, regulates vacuolar fusion, autophagy and plant infection (Li et al, ). Rab7‐mediated FgAtg9 trafficking is essential for autophagy‐dependent development and pathogenicity in F. graminearum (Zheng et al, ).…”
