Fibrates ameliorate the course of bacterial sepsis by promoting neutrophil recruitment via CXCR2

Abstract: Bacterial sepsis results in high mortality rates, and new therapeutics to control infection are urgently needed. Here, we investigate the therapeutic potential of fibrates in the treatment of bacterial sepsis and examine their effects on innate immunity. Fibrates significantly improved the survival from sepsis in mice infected with Salmonella typhimurium, which was paralleled by markedly increased neutrophil influx to the site of infection resulting in rapid clearance of invading bacteria. As a consequence of … Show more

“…Chemotaxis of neutrophils was measured with 48-well micro-chambers and polycarbonate filters (5 μm pore size) (NeuroProbe, Cabin John, MD) as described ( Tancevski et al, 2014 ). The results were expressed as the mean ± SEM of the chemotaxis index (CI), representing the fold increase in the number of migrated cells in response to chemo-attractants over spontaneous cell migration (to control medium).…”

“…Even though chemotactic pathways were not specifically investigated, cytokines, cytokine receptors, and related proteins were included per our computational criteria in our construction of a drug’s activity network and anthrax toxins’ activity networks in macrophages. Interestingly, fenofibrate reportedly preserved expression of CXCR2 on neutrophils and facilitated neutrophil migration, offering protection in sepsis [76]. Simvastatin inhibited IL-5-induced chemotaxis of human eosinophils [77] and reduced C-reactive protein-induced chemotaxis in human monocytes [78].…”

“…There are multiple other agents being studied at present in preclinical models targeting a wide range of inflammatory, immune, metabolic, bioenergetic, and hormonal pathways (Dalli et al , ; Tancevski et al , ; Winkler et al , ; Rathkey et al , ; Sham et al , ). Transfer of mesenchymal stem cells appeared promising in preclinical studies, and phase 1 and 2 clinical trials are ongoing (Keane et al , ).…”
