Fibroblast‐mediated intercellular crosstalk in the healthy and diseased heart

Abstract: Cardiac fibroblasts constitute a major cell population in the heart. They secrete extracellular matrix components and various other factors shaping the microenvironment of the heart. In silico analysis of intercellular communication based on single‐cell RNA sequencing revealed that fibroblasts are the source of the majority of outgoing signals to other cell types. This observation suggests that fibroblasts play key roles in orchestrating cellular interactions that maintain organ homeostasis but that can also c… Show more

“…In support of this, Fgf2 mRNA expression was significantly upregulated in hearts of mice treated with AngII, and the increase in expression was reduced in mouse hearts with cardiomyocyte BRAF knockout (Figure 3D). This is consistent with the concept of cross-talk between cardiomyocytes and the non-myocytes in the heart [36].…”

“…Among the three isoforms, TGF-β1 is predominant. It is crucial in pathological fibrosis and is produced by various cells, including immune cells, endothelial cells, cardiomyocytes, and activated fibroblasts [ 111 , 112 ]. TGF-β1 is initially secreted as an inactive complex with latent TGF-β-binding proteins and TGF-β pro-peptides.…”

“…Andreas Herrlich’s review ‘Interorgan crosstalk mechanisms in disease: the case of acute kidney injury‐induced remote lung injury’ [41] highlights the role of kidney‐released mediators that cause lung endothelial leakage, lung edema, lung inflammation, and respiratory failure as one important example of such clinically important crosstalk. Luka Nicin, Julian U G Wagner, Guillermo Luxán and Stefanie Dimmeler’s review ‘Fibroblast‐mediated intercellular crosstalk in the healthy and diseased heart’ [42] reports current knowledge of cellular interactions in the healthy, diseased, and aging heart and their potential relevance for interorgan crosstalk.…”
