Fibronectin Enhances In Vitro Lipopolysaccharide Priming of Polymorphonuclear Leukocytes

Abstract: We investigated the role of humoral factors in lipopolysaccharide (LPS) priming of polymorphonuclear leukocytes (PMN) using cells isolated from adults and from neonates. Plasma from newborn infants had decreased priming activity of adult plasma when mixed with LPS in studies measuring oxidative radical production of PMN after stimulation with a formyl bacterial oligopeptide (fMLP). This marked difference was not caused by LPS binding protein (LBP) because the LBP concentration in newborn and adult plasma were … Show more

“…Activated monocytes liberate early the cytokines TNF‐α and interleukin (IL)‐1 and, together with other factors (13), prime and sensitize neutrophils to the effects of other agonists. Preoperative immune activation may be due to congenital heart disease (14, 15) or to subclinical inflammation (9), or it may result from an atopic/allergic predisposition (16, 17).…”

“…It is desirable to predict postoperative complications, induced by an activated immune system (1, 9, 13), from a predisposition by the cardiac disease (14, 15) or from other causes (e.g., atopy/allergy; 16,17) already present preoperatively (7). Predisposed patients may respond more dramatically to CPB and surgical trauma.…”

“…hyaluronan secreted more cytokine than cells treated with LPS alone [106]. Fibrinogen and fibronectin were shown to prime neutrophils toward greater responsiveness to LPS [108,104], and fibronectin was shown additionally to enhance microglial cell responses to lipopeptide and CpG DNA [105]. Mice genetically deficient in biglycan were shown to be considerably less responsive to LPS than wildtype animals [35] in a manner similar to that shown for the related LRR-containing ECM component lumican [97].…”
