Fidelity of synthesis of preribosomal RNA in isolated nucleoli and nucleolar chromatin.

Abstract: Previously, other workers have shown that isolated nucleoli can synthesize RNA in vitro that resembles pre-rRNA in base composition, marker oligonucleotides, and hybridization analysis (4-10). The present study further supports the fidelity of pre-rRNA synthesis by isolated nucleoli and nucleolar chromatin, using the homochromatographic fingerprinting technique, which permits a direct comparison of patterns of virtually all the oligonucleotides obtained by T1 ribonuclease digestion of 45S pre-rRNA.Abbreviation… Show more

“…RNA synthesis in isolated Physarum nuclei maintains a high degree of fidelity with regard to transcription of rDNA coding regions and exclusion of most of the central spacer region. Using a variety of criteria for evaluation, several workers have reported the fidelity of rDNA transcription in the nuclei of several organisms (Zylber & Penman, 1971;Reeder & Roeder, 1972;Ballal et al, 1977). Upon hybridizing either total labeled nuclear RNA or HS-nRNA to Hpal restriction fragments (Figure 4), it can be seen that fragments of 5.9 and 3.2 kb, located within the central spacer, are not transcribed.…”

“…The appearance of a similar number of spots displayed in the same characteristic fashion on the homochromatogram (Fig. 2) shows that the RNA transcribed in vitro closely resembles the RNA labeled in vivo (2). This technique differentiates specific and random transcripts inasmuch as the transcripts obtained from nucleolar chromatin in Fig.…”

“…In the nucleolus, tandemly repeated 45S ribosomal RNA genes (or rDNA) are transcribed to form 45S nascent pre-rRNA (Ballal et al, 1977). Each prerRNA is cleaved and processed to form the 18S, 5.8S and 25S rRNA molecules which are essential for the formation of ribosomes (Shaw and Jordan, 1995).…”
