Fine Mapping of 28S rRNA Sites Specifically Cleaved in Cells Undergoing Apoptosis

Abstract: Bona fide apoptosis in rat and human leukemia cells, rat thymocytes, and bovine endothelial cells was accompanied by limited and specific cleavage of polysome-associated and monosome-associated 28S rRNA, with 18S rRNA being spared. Specific 28S rRNA cleavage was observed in all instances of apoptotic death accompanied by internucleosomal DNA fragmentation, with cleavage of 28S rRNA and of DNA being linked temporally. This indicates that 28S rRNA fragmentation may be as general a feature of apoptosis as internu… Show more

“…The block of protein synthesis by cycloheximide (CHX) during the early phase of cAMP-induction confers resistance to cAMP-induced cell death, as shown by inhibition of DNA fragmentation (Lanotte et al, 1991), 28S rRNA cleavage (Houge et al, 1993(Houge et al, , 1995 and in this paper, Figure 5a (lower panel), compare lanes 4 ± 6 (without CHX) and lanes 9 ± 11 (with CHX). In these experiments CHX treatment alone is not inducing rRNA cleavage (Figure 5a Figure 5c, after 3 h).…”

“…Furthermore, cell death is delayed by treating IPC-81 cells with actinomycin D or cycloheximide. This treatment suppresses morphological changes typical of apoptosis, as well as DNA fragmentation and 28S rRNA cleavage (Lanotte et al, 1991;Houge et al, 1993Houge et al, , 1995. This suggest a direct involvement of PKA-directed gene expression.…”
