First identification of porcine parvovirus 6 in Poland

Abstract: Porcine parvovirus type 1 is a major causative agent of swine reproductive failure. During the past decade, several new parvoviruses have been discovered in pigs. Porcine parvovirus type 6 (PPV6), recently identified, has been reported in pigs in China and in the USA while the PPV6 status in the European pig population remains undetermined. In the present study, PPV6 DNA was identified in serum samples collected from domestic pigs in Poland. In investigated herds, the prevalence of PPV6 was 14.9 % (15/101 samp… Show more

“…PPV5 and PPV6 represent very recently identified PPV genotypes in China and the USA (Xiao et al., ; Ni et al., ; Wu et al., ; Schirtzinger et al., ), but knowledge of their circulating in Europe is still limited (Cui et al., ; Fan et al., ). In Poland, the overall prevalence rate of PPV5 was only 4.0%.…”

“…Duplex (PPV1 and PPV2) and triplex (PPV3, PPV4 and PPV5) differential real‐time PCR assays were performed as described (Opriessnig et al., ). In addition, a real‐time PCR assay for PPV6 was performed as described (Cui et al., ). Amplifications were carried out on a 7500 Fast Real‐time PCR system (Applied Biosystems) under the following conditions: initial denaturation at 95°C for 2 min, followed by 40 cycles of 95°C for 15 s and 60°C for 30 s. Samples with cycle threshold (Ct) values over 37 were considered negative.…”

“…Three duplex real-time PCR assays were performed to detect PPV1 and PPV2, PPV3 and PPV6, PPV4 and PPV5 using previously published primers and probes [9,32,33,35,36] (Table S2). The amplification was carried out on 6000 Rotor Gene (Qiagen, Hilden, Germany) and each reaction of a total volume of 25.0 µL contained 12.5 µL of a reaction mix (SensiFAST Probe No-ROX, Bioline), 2.0 µL of DNA, 1.0 µL of each 10.0 µM primer, and 0.5 µL of each 10.0 µM probe.…”

“…PPV6 was first isolated in Chinain 2014 [8] and similar to PPV4 [13,14]. Furthermore, PPV7 was isolated in China since 2017.…”
