First Molecular Detection and Vp7 (G) Genotyping of Group a Rotavirus by Semi-Nested Rt-PCR From Sewage in Nigeria

Abstract: SUMMARYRotavirus is the leading cause of viral gastroenteritis worldwide, and sewage is a major source of the virus dissemination in the environment. Our aim was to detect and genotype rotaviruses from sewages in Nigeria. One hundred and ninety sewage samples were collected between June 2014 and January 2015. The two phase concentration method using PEG 6000 and dextran was used to concentrate sewage samples following WHO protocols. Molecular detection was performed by RT-PCR, and VP7 genotyping by semi-nested… Show more

“…During this time seven VP7 genotypes were reported from Nigeria, genotypes G1, G2, G3, G8, G9, G10, and G12, with genotypes G9 and G12 being the most recent isolates [ 22 – 24 , 26 29 ]. Environmental isolates recovered from sewage effluent were also identified in our analysis [ 42 ]. Our analysis shows a rapid emergence and dominance of genotype G12 lineage III in Nigeria.…”

“…Some articles assessed the presence of poliovirus, [69][70][71][72] either wildtype or vaccine-derived, including potential neurovirulence of vaccine-derived poliovirus. [73][74][75] Many articles documented the diversity of non-polio enteroviruses found in wastewater, 73,[76][77][78][79][80][81][82][83][84][85][86][87][88][89][90][91][92] with a variety of focuses including rotavirus, [93][94][95][96][97] norovirus, 88,92,[98][99][100] astrovirus, 101,102 polyomavirus, 103 Saffold virus, 104 hepatitis A virus, 85 hepatitis E virus, 105 mastadenovirus, 106 Aichi virus, 107 and human bocavirus. 107 Surveillance of Giardia and/or Cryptosporidium was also documented.…”

“…However, rotavirus still causes about 192,700 deaths per year in children ˂5 years mostly in low-and middle-income countries [6]. In Nigeria, they have been reports of various genotypes including mixed and unusual genotype combination isolated from young children and environmental samples [7,8,9]. This might have undermined the public health efforts in controlling pediatric rotavirus infection in Nigeria.…”
