First Prize: Isoprostane: Quantitation of Renal Ischemia and Reperfusion Injury After Renal Artery Clamping in an Animal Model

Abstract: Our data have demonstrated that isoprostane levels are a potential real-time marker of renal ischemia and reperfusion injury. We also found allopurinol administration demonstrated a trend toward renoprotective abilities in the hilar occluded kidney.

“…One of the approaches used to quantify oxidative stress in vivo consists of measuring the 8-isoprostane levels. In Wang et al 11 study, such levels significantly increased after 60 minutes of only ischaemia, while they did not change significantly after 15, 30 and 45 minutes. In that study, the authors compared the 8-isoprostane levels between two groups of rats subjected to 30 minutes of reperfusion after a period of 60 minutes of renal ischaemia, one of which was given 100 mg·kg -1 intravenous allopurinol, while the other group received no treatment.…”

“…Warm ischemia was identified as the “ultimate enemy” for partial nephrectomy [ 20 ], and several methods to ameliorate its negative aspects have been proposed [ 5 8 ]. Although mannitol is largely employed for this purpose [ 9 ], its effects for ischemia protection during partial nephrectomy were only recently investigated [ 10 , 11 ].…”

“…Furthermore, recent studies have shown that the quality and quantity of remnant renal parenchyma is of great importance to predict renal function [ 4 ]. Thus, various methods have been employed to prevent damage to the remnant kidney parenchyma after prolonged warm renal ischemia [ 5 8 ].…”

“…Thus, here the applicability of the trans/cis-epoxy-PUFA ratio as oxidative stress marker cannot be evaluated based on comparison with IsoP levels. Even though IRI is linked to excess ROS formation [50,51] and an increase of oxidative damage derived IsoP following renal IRI has been observed in rat models [52,53], there are various factors influencing measured IsoP levels in this model. These include clamping time, since longer clamping seem to lead to higher IsoP levels [52], as well as the time point of sampling following reperfusion and the specimen analyzed, e.g.…”
