Fisetin Suppresses the Inflammatory Response and Oxidative Stress in Bronchial Epithelial Cells

Abstract: Fisetin is isolated from many fruits and vegetables and has been confirmed to improve airway hyperresponsiveness in asthmatic mice. However, whether fisetin reduces inflammatory response and oxidative stress in bronchial epithelial cells is unclear. Here, BEAS-2B human bronchial epithelial cells were treated with various concentrations of fisetin and then stimulated with tumor necrosis factor-α (TNF-α) or TNF-α/interleukin-4. In addition, ovalbumin-sensitized mice were treated with fisetin to detect inflammato… Show more

“…Fisetin, chemically 3,7,3′,4′-tetrahydroxyflavone or 7,3′,4′-flavon-3-ol is a polyhydroxy flavonoid with various pharmacological potentials like antioxidant ( Khan et al, 2013 ), anti-inflammatory ( Wu et al, 2022 ), anti-cancer ( Lin et al, 2016 ), neuroprotective ( Kumar et al, 2020 ) and is used in vascular dementia ( Cordaro et al, 2022 ). It occurs in numerous fruits and vegetables, such as grapes, apples, onions, strawberries, and cucumbers ( Cordaro et al, 2022 ).…”

“…At 40 mg/mL methacholine, asthmatic mice treated with gypenoside A or prednisolone had significantly reduced Penh values when compared with the OVA group mice ( Figure 2 A). Mice were anesthetized and bronchoalveolar lavage fluid (BALF) was collected as described previously [ 16 ]. Compared with the OVA group mice, asthmatic mice treated with prednisolone or gypenoside A also had significantly decreased numbers of eosinophils in BALF ( Figure 2 B).…”

“…In this study, the drug concentrations in animal experiments were selected with reference to the published similar literature (Shanmugam et al, 2018; Wu et al, 2022). Also, the concentration of fisetin was selected by referring to 1/10th of the LD50 concentration in the literature (Hassan et al, 2022).…”

“…The dose of DDP was chosen with reference to similar published literature (Gouttia et al, 2022; H. Li, Huang, Yu, & Lou, 2020), while the dose of fisetin was chosen with reference to the LD50 (median lethal dose) value of fisetin by intraperitoneal injection in mice (250 mg/kg; Hassan et al, 2022), in addition to the doses chosen in other in vivo experiments with fisetin (Shanmugam, Ravindran, Kurian, & Rajesh, 2018; Wu et al, 2022), and was chosen not to exceed 1/10 of the LD50 value.…”
