Fission yeast homologues of the B′ subunit of protein phosphatase 2A: multiple roles in mitotic cell division and functional interaction with calcineurin

Abstract: Background: Protein phosphatase 2A (PP2A) is a serine/threonine phosphatase distributed in eukaryotes from yeast to human, and plays pivotal roles in diverse cellular functions such as metabolism, cell cycle progression, gene expression and development. PP2A holoenzyme is a heterodimer of a catalytic subunit C and a regulatory subunit A, or a heterotrimer of C, A and a variable regulatory subunit consisting of three families; B, B 0 , and PR72. Specific functions for each variable subunit are not well understo… Show more

“…At the same time, S. pombe pab1-4 cells lose growth polarity and manifest disorganized microtubule arrays and delocalized cortical actin patches ( Figure 6A), a phenotype in agreement with previous studies of the pab1 null allele, which established a positive role for PP2A-Pab1 in morphogenesis (Kinoshita et al 1996;Jiang and Hallberg 2000;Tanabe et al 2001). Thus, PP2A-Pab1 seems to play a dual function: negative regulation of cell division with a positive role in morphogenesis.…”

“…In S. pombe, there are two genes encoding the B9-regulatory subunits, par1 and par2. The double null mutant is viable, but is sensitive to temperature (high and low) and stress and shows defects in septation and cleavage ( Jiang and Hallberg 2000;Tanabe et al 2001). Curiously, a mutation in the regulatory subunit B9 Par1 is able to suppress the septation defects of cdc11-136, cdc7-24, spg1-B8 (Le Goff et al 2001), and sid2-250 mutants ( Jin et al 2006).…”

“…The phenotype of the pab1-D mutant suggests that this form of PP2A is important for cell morphogenesis (Kinoshita et al 1996;Tanabe et al 2001;Lahoz et al 2010). The similarity of this to the ppa2-6 and ypa2-D indicates that PP2A plays an important role in cell morphogenesis and also implicates Ypa2p in this process.…”

“…The par1 and par2 genes encode the two B9-family subunits. Par1p is the major form (Jiang and Hallberg 2000;Le Goff et al 2001;Tanabe et al 2001); deletion of par1 affects septum positioning and also renders cells cold sensitive and hypersensitive to loss of PP2B/calcineurin (Jiang and Hallberg 2000;Le Goff et al 2001;Tanabe et al 2001); in contrast, deletion of par2 has little effect on the cell per se (Jiang and Hallberg 2000;Tanabe et al 2001).…”

“…Protein phosphatases Par1p and Par2p all act negatively on septation as deletion of par1 and par2 genes leads to a phenotype with multiple septa formed between daughter cells ( Jiang and Hallberg 2000;Tanabe et al 2001). The par1 gene also was isolated as an extragenic suppressor of loss-of-function mutation in the spg1 gene (Le Goff et al 2001).…”
