FITC‐labeled lipopolysaccharide: use as a probe for liposomal membrane incorporation studies

Abstract: FITC-labeled LPS from Neisseria meningitidis can be used as a probe to follow the process of LPS incorporation into liposomal membrane and to study its interaction with a bilayer. The incorporation of FITC-LPS into the bilayer was proved by physicochemical methods as well as by liposomal LPS toxicity decrease in actinomycin D-sensitized mice. Fluorescence intensity increase was observed upon the insertion of FITC-LPS into the membrane of dehydration/rehydration vesicles and vesicles obtained by co-sonication o… Show more

“…6). In agreement with our results, LPS clustering was previously observed upon incorporation of LPS isolated from group B Neisseria meningitidis strain B125 in mixtures of egg PC/cholesterol at low LPS concentrations (39). Taking into account the aforementioned results, we can hypothesize that the clustering ability of LPS at low concentrations would also be observed in compositional environments that are representative of both mammalian and bacterial membranes.…”

“…Magainin 2, as well as other cationic antimicrobial peptides, is known to only weakly interact with the host zwitterionic lipid [19,21]. LPS is reported to form clusters in a phospholipid matrix [22]. Therefore, we can extract specific interactions between the peptide and the LPS domain.…”

“…Acid form and salts of LPS. The Na+ salt of LPS, sufficiently soluble in water, was obtained after chromatography on a column packed with Sephadex G-200 in a buffer solution of EDTA and sodium deoxycholate (27). At this stage, LPS was separated from the high-molecular-weight components.…”

“…The incorporation of LPS into liposomes was performed by the method of dehydration-rehydration of vesicles (DRV) and the method of prolonged cosonication (25,27). In the DRV method, 150 ,ul of pyrogen-free deionized water was added to a lipid film, well dried from the traces of organic solvents, and the lipid was resuspended by vigorous vortexing.…”
