Flavonoids from the leaves of Actinidia kolomikta

Abstract: Actinidia kolomikta (Rupr. et Maxim.) Planch. The chemical structures of the isolated compounds were established by application of chemical and spectroscopic analysis. The new acetylated flavonoid was screened for its protective effect on human erythrocytes against AAPH-induced hemolysis, and it can slow the hemolysis induced by AAPH.

“…[10] Comparing with the reported data, the 1 H NMR and 13 C NMR data are in agreement with those of Isoorientin and Isoorientin 6″-O-acetate in the literature. [8][9][10][11] Compound 3: 3,4-dihydro-6,8-dihydroxy-3-methylisocoumarin (6-hydroxymellein) Figure 3 [4] Analytical CPG/SM of the ethyl acetate extract of Retura lutea indicated the presence of three major volatile compounds. The major constituent which eluted at a lower temperature was identified as 3,4-dihydro-6, 8-dihydroxy-3-methylisocoumarin) with a retention time of 18.270 min (Figure 4).…”

“…Hemolysis induced by AAPH [2,2'-azobis(2-methylpropionamidine) dihydrochloride] was determined by the method of Lu et al . [55] with the following modifications. The erythrocytes were washed threes time with phosphate buffer saline (PBS) pH 7.4, then a suspension of human erythrocyte in (5%) PBS was prepared.…”

“…To perform anti-hemolytic activity, firstly the hemolysis was induced by AAPH (2,2′-azobis-[2-methylpropionamidine]) radical according to the methodology of Lu et al (2010) with some modifications. The globular package was separated from the plasma by centrifugation at 2000 × g for 10 min at 4 °C.…”

“…The supernatant was read at 540 nm on a 96-well microplate. The half maximal-inhibitory concentration (IC 50 ) was determined following Lu et al (2010) procedure. The percentage of hemolysis inhibition (% HI) was obtained from the equation (5) : where AAPH 1 is the optical density of the hemolysis caused per radical AAPH, and HS is the optical density of the hemolysis inhibition by each treatment.…”
