Flow Cytometric Analysis of Leukocytes in the Human Female Reproductive Tract: Comparison of Fallopian Tube, Uterus, Cervix, and Vagina

Abstract: Three-color flow cytometry is an appropriate method for quantitative comparison of leukocytes from the different tissues of the female reproductive tract, during all phases of the menstrual cycle and within post-menopausal samples. Results indicate that the tissues differ from each other, particularly with respect to the large number of granulocytes in the Fallopian tubes.

“…NK cells appear during the proliferative phase and increase in cell numbers during the secretory phase of the menstrual cycle (12)(13)(14). A portion of this expansion is most likely due to proliferation of NK cells, because some are found in late secretory phase that express the Ki-67 Ag indicative of cell proliferation (29).…”

“…The interplay between these different cell types and their roles in defense against pathogen invasion in this specialized tissue are poorly understood. Unlike in the murine uterus, uNK cells in the human uterus are found widely dispersed throughout the EM, and their numbers increase as the menstrual cycle progresses (12)(13)(14).…”

“…With menopause, sex hormone fluctuations decline and hormone levels remain low and constant (Givan et al., 1997; Wira, Rodriguez‐Garcia & Patel, 2015). Sex hormones regulate immune cell populations and function in the FRT throughout the menstrual cycle to prepare for implantation and pregnancy (Wira et al., 2015).…”

“…For instance, epithelia and stromal cells of the reproductive organs of the mouse and human, express high levels of TGF-1 and other molecules involved in conditioning immune privilege sites (Chegini et al, 1994;Grant and Wira, 2003;Jin et al, 1992;Wada et al, 1996). Moreover, antigen presenting cells such as macrophages and DCs, regularly present in the reproductive tissues might also contribute to regulatory response, since they could promote T regulatory cells differentiation trough production of IL-10 and TGF-1 after infection (Givan et al, 1997;Stagg et al, 1998).…”
