Pathogen infection poses a serious threat to the survival and commercial quality of cultured Paa spinosa, which provide protection as a substitute for wild P. spinosa. The gut microbiota plays vital roles in host health and immunity. To provide guidance for preventing intestinal diseases of artificial P. spinosa culture, we compared gut microbiota compositions of wild and cultured P. spinosa using high‐throughput sequencing. A total of 11,526 operational taxonomic units (OTUs) were identified from 14,043 sequences from each sample. Cetobacterium, PW3 Bacteroides and some unidentified species from Bacteroidales, Rikenellaceae and Clostridiales were significantly increased in the gut microbiota from cultured P. spinosa, whereas Faecalibacterium and unidentified bacteria from Ruminococcaceae were significantly decreased in the gut microbiota from cultured P. spinosa. According to the gut microbiota composition, we hypothesized that the cultured P. spinosa in Jing'an would exhibit a higher risk of pathogenic infection than those in Cili. These results provided a method to forecast the pathogenic infection risk of cultured P. spinosa, which could guide the artificial culture of and prevent diseases in P. spinosa through gut microbiota.

rEsumoO controle químico de plantas daninhas é um método eficaz utilizado em florestas plantadas e em áreas de restauração florestal, favorecendo o desenvolvimento das mudas florestais e a redução dos custos de intervenção. Entretanto, se essa estratégia não for aplicada de maneira correta, pode-se tornar um potencial causador de impactos negativos aos organismos do solo e aos processos ecológicos que esses participam. avaliou-se o efeito do uso de herbicidas na biota do solo por meio da amostragem da comunidade da fauna edáfica e testes ecotoxicológicos de reprodução e sobrevivência de Folsomia candida e Enchytraeus crypticus, em área-alvo de restauração florestal recém-implantada. O delineamento experimental adotado foi de blocos casualizados com quatro tratamentos e cinco repetições. os tratamentos consistiram da aplicação dos herbicidas pós-emergentes mesotrione (0,4 l ha -1 ), fluazifop-P-butyl (1,0 L ha -1 ) e nicosulfuron (1,5 l ha -1 ), aplicados em área total nas parcelas, mais o controle (sem aplicação). na área de estudo, foram realizadas duas aplicações dos herbicidas com intervalos de seis meses, onde as parcelas foram correspondentes aos tratamentos em ambas as aplicações. Em cada parcela, foram realizadas quatro coletas de solo, sendo a primeira (coleta 1) seis dias antes da segunda aplicação dos herbicidas na área. as demais, após um (coleta 2), oito (coleta 3) e 22 dias (coleta 4) dessa mesma aplicação. a comunidade da fauna do solo foi avaliada por meio do método de funis de Berlese modificado, utilizando-se uma amostra de solo por parcela. Em laboratório, os indivíduos foram contados e identificados em grandes grupos. Os ensaios ecotoxicológicos de sobrevivência e reprodução de F. candida e E. crypticus foram realizados a Recebido para publicação em 15 de maio de 2015 e aprovado em 28 de julho de 2015.

Ultra‐performance convergence chromatography with tandem mass spectrometry‐assisted method for rapid enantioseparation and determination of fluazifop‐butyl in tobacco and soil

A quick, green, and sensitive method for chiral separation and determination of fluazifop-butyl enantiomers in tobacco and soil was established by ultraperformance convergence chromatography with tandem mass spectrometry (UPC 2 -MS/MS). The baseline separation was obtained on an ACQUITY UPC 2 Trefoil CEL2 column in 4 minutes with CO 2 and methanol as mobile phase. Column temperature, auto back pressure regulator pressure (ABPR), and modifier solvent were optimized to obtain the best separation efficiency.Under the optimal conditions, the recoveries of both enantiomers were 82.8% to 99.5% with relative standard deviations (RSDs) less than 5.5% at three different concentration levels in two matrices. Good coefficients of determination (R 2 ≥ 0.9976) were achieved over the concentration range of 10 to 500 ng/mL. The limits of detection (LODs) for all enantiomers in the two matrices varied from 1.6 to 2.1 μg/kg, and the limits of quantification (LOQs) did not exceed 7.0 μg/kg. The proposed method was then successfully applied to analyze authentic samples, confirming that it was a green, convenient, and reliable strategy for the analysis of fluazifop-butyl enantiomers in tobacco and soil.
