Fluid dynamics in zebrafish Kupffer's vesicle

Abstract: Work in mouse has implicated cilia motility and leftward nodal flow as the mechanism for breaking left-right symmetry. In zebrafish, it is assumed that Kupffer's vesicle is analogous to the mouse node.

“…The KV is a transient fluid filled organ derived from the dorsal forerunner cells that plays a critical role in establishing L/R polarity (Cooper and D’Amico, 1996; Essner et al, 2005; Kramer-Zucker et al, 2005; Melby et al, 1996; Okabe et al, 2008). We have generated a transgenic reporter line Tg(dusp6:d2EGFP) pt6 that expresses GFP in response to FGF signaling (Molina et al, 2007).…”

“…In zebrafish, the cellular architecture of KV is asymmetric along the anteroposterior and dorsoventral axes. KV cells in the dorsoanterior region develop an elongated columnar morphology with small apical surfaces-which allows tight packing of these cells-whereas ventroposterior cells adopt a cuboidal morphology with large apical surfaces [37,38]. This process, referred to as 'KV remodelling' [72], places more motile cilia in the dorsoanterior region to drive strong leftward flow across the anterior pole as described above.…”

“…In addition, Ca 2+ oscillation in the cilia is thought to regulate left-right asymmetry 42)43)44). Although the cilia of the KV are motile,20) they are proposed to express PKD2 and sense extracellular Ca 2+ , as the immotile cilia in mouse perinodal crown cells do, although the cilia have 9+2 type microtubules in the KV 4). The movement of cilia and intracellular Ca 2+ can be monitored by the small GTPase, Arl13b, tagged with genetically encoded Ca 2+ indicators, GCaMP5 or RGECO1 42).…”

“…The dorsal forerunner cells (DFCs) of the endoderm give rise to KV-forming cells 18)19). Subsequently, these cells become ciliated to promote unidirectional flow in the KV 20). Motile cilia rotate unidirectionally from right to left to induce unidirectional flow in the KV 21).…”
