Fluorescence Identification of Head and Neck Squamous Cell Carcinoma and High-Risk Oral Dysplasia With BLZ-100, a Chlorotoxin-Indocyanine Green Conjugate

Abstract: Importance Surgical cure of head and neck squamous cell carcinoma (HNSCC) remains hampered by inadequately resected tumors and poor recognition of lesions with malignant potential. BLZ-100 is chlorotoxin-based tumor targeting agent which has not yet been studied in HNSCC. Objective To evaluate BLZ-100 uptake in models of HNSCC and oral dysplasia. Design Observational study (including sensitivity/specificity analysis) of BLZ-100 uptake in an orthotopic xenograft mouse model of HNSCC and a carcinogen-induced… Show more

“…DISCUSSION Tozuleristide, in preclinical studies, has been shown to bind in both primary and metastatic disease to a variety of tumor types including gliomas, sarcomas, and head and neck squamous cell carcinomas. 6,[8][9][10] Here we show for the first time that tozuleristide binds breast carcinomas in a clinical study. Signal for invasive carcinoma appeared bright and focal in ex vivo imaging of sectioned fresh breast tissue.…”

“…Thus, BLZ-100 is a highaffinity, targeted fluorescent probe that has shown specificity for glioma cells and is capable of near-infrared fluorescence, making it viable for applications of fluorescence-guided glioma resection. BLZ-100 has seen success in preclinical studies for resection of glioma (120), head and neck carcinoma (121), and soft-tissue sarcoma (122). Liposomes targeted with CTX were also tested in mouse glioblastoma (283).…”
