Fluorescence Imaging During Photodynamic Therapy of Experimental Tumors in Mice Sensitized with Disulfonated Aluminum Phthalocyanine¶

Abstract: A fluorescence imaging system was used to monitor the emission of disulfonated aluminum phthalocyanine (AlS2Pc) during the photodynamic therapy (PDT) of murine tumors. Cells of the MS‐2 fibrosarcoma were injected in mice in two compartments in order to cause the development of tumors in different host tissues. Two drug doses and two uptake times were considered. Moreover, the fluorescence of the AlS2Pc was excited using two wavelengths on the opposite sides of the absorption peak to detect a possible change in… Show more

“…This represents much greater light‐induced photodegradation of Pc 4 than that observed in the clinical study (32) where approximately 53% of the fluorescence was lost after irradiation. We observed greater photobleaching of Pc 4 than the photobleaching of AlS 2 Pc observed following laser irradiation of AlS 2 Pc‐treated tumors (44). The differences in the extent of photobleaching in these studies may be due to the different tumor types evaluated and differences in the microvasculature of the tumors.…”

“…In mice receiving AlS 2 Pc‐PDT at 12 h after AlS 2 Pc was administered, AlS 2 Pc photobleaching was observed. However, when the time between dosing and laser irradiation was 3 h, increased fluorescence was observed immediately after laser irradiation (44). Cubeddu et al.…”

“…In vivo a similar transient intensity rise followed by photobleaching has been observed by us in the liver of anaesthetised AlS 2 Pc-sensitised rats monitored under laparotomy (unpublished data) and by others in tumour models. 24,25 In the study by Cubeddu et al 24 a variable response was noted between individual tumours some of which exhibited little fluorescence increase, which was attributed to variation in tumour uptake. Likewise in the present study some cells in one field of view did not exhibit fluorescence enhancement, which is most likely due to variable patterns of uptake between cells.…”
