Fluorescence properties of several chemotherapy drugs: doxorubicin, paclitaxel and bleomycin

Abstract: Several chemo-drugs act as the biocompatible fluorophores. Here, the laser induced fluorescence (LIF) properties of doxorubicin, paclitaxel and bleomycin are investigated. The absorption lines mostly lie over UV range according to the UV-VIS spectra. Therefore, a single XeCl laser provokes the desired transitions of the chemo-drugs of interest at 308 nm. It is shown that LIF spectra are strongly dependent on the fluorophore concentration giving rise to the sensible red shift. This happens when large overlappin… Show more

“…if the observed downregulation of ABCB1 expression after ROR1 inhibition correlated with a decrease in chemo drug efflux, we monitored doxorubicin in cells following ROR1 inhibition. Doxorubicin has intrinsic fluorescence 29 and can be monitored via confocal microscopy. We observed an increase in doxorubicin levels within cells 24 h after treatment, following ROR1 knockdown and Strc inhibition, when compared to control RNA and vehicle treatment groups ( Fig.…”

“…The resulting blanked spectrum overlaid with PTX and FA spectra is shown in Figure c. The shoulder peak at 227 and 272 nm are characteristic peaks of PTX and FA, respectively, confirming the presence of both the molecules 8a,21. The amount of PTX and FA was quantified by using Beer's law, and the result shows the linear relationship between absorbance and concentration of PTX for the concentration range of 5–20 µg mL −1 (Figure S1a,b, Supporting Information); a linear relationship was also found between the absorbance and concentration of FA at the concentration range of 1.25–5 µg mL −1 (Figure S2b, Supporting Information); approximately 173 PTX molecules and 82 FA molecules per NP were detected.…”

“…For the synthesis of fluorescently labelled nanogels (Nanogel Cy5 ), 2% of the reactive esters on the polymer were substituted with cyanine5-amine prior to crosslinking. We chose cyanine5 as a labelling agent because its fluorescence excitation and emission spectra do not, contrary to rhodamine and fluorescein, show overlap with the spectra of doxorubicin 52 .…”
