Fluorescent risedronate analogues reveal bisphosphonate uptake by bone marrow monocytes and localization around osteocytes in vivo

Abstract: Bisphosphonates are effective antiresorptive agents owing to their bone-targeting property and ability to inhibit osteoclasts. It remains unclear, however, whether any non-osteoclast cells are directly affected by these drugs in vivo. Two fluorescent risedronate analogues, carboxyfluorescein-labeled risedronate (FAM-RIS) and Alexa Fluor 647–labeled risedronate (AF647-RIS), were used to address this question. Twenty-four hours after injection into 3-month-old mice, fluorescent risedronate analogues were bound t… Show more

“…It was reported that cells in soft tissues take up BPs from circulation, and suffer apoptosis or impairment of their functions. 54 In our work, the alendronate was strongly bound to the CaP coating on Ti. Thus, it has the advantage of only directly affecting the area surrounding the implant, which may reduce the above-mentioned adverse side effects of BPs.…”

“…(1) Very recently it was shown that another bisphosphonate (risedronate) is taken up by bone marrow monocytes and localized around osteocytes in vivo and that there is a direct interaction between these osteocytes and the bisphosphonate. (36) Whether such is the case with ZOL remains to be determined. Despite the fact that further experiments are needed to decipher the exact mechanisms leading to the current observations, these results offer a glimpse into potential additional mechanisms by which bisphosphonates may exert their beneficial antifracture effect during osteoporosis treatment.…”
