The 2 most abundant polycyclic aromatic hydrocarbons (PAHs) measured in Deepwater Horizon crude oil, naphthalene and phenanthrene, and their associated homologs have both been shown to be acutely toxic in fish. Although fish have a relatively high metabolic capacity for PAHs, hydroxylated PAH (OH-PAH) derivatives formed during the initial metabolic response can negatively impact the health of fish. Species-specific metabolism of naphthalene and phenanthrene was evaluated in 3 marine teleosts, red drum (Scianops ocellatus), Florida pompano (Trachinotus carolinus), and southern flounder (Paralichthys lethostigma). Fish were exposed to Deepwater Horizon crude oil by intraperitoneal injections at time 0 and 48 h, with bile sampling events at 24 and 72 h post injection. The data suggested metabolic induction in Florida pompano and red drum, whereas southern flounder may have demonstrated metabolic fatigue. By 24 h post injection, overall profiles of red drum and southern flounder were dominated by hydroxylated phenanthrene metabolites; conversely, the Florida pompano profiles were dominated by monohydroxylated naphthalenes. In addition, Florida pompano had faster overall relative biotransformation rates, suggesting their potential decreased susceptibility to adverse effects. Red drum and southern flounder had much lower relative biotransformation rates, indicating their probable susceptibility to adverse outcomes after naphthalene and phenanthrene exposures. To our knowledge, the present study is the first to investigate monohydroxylated PAHs in fish exposed to Deepwater Horizon oil.

Early life stages of fish are highly sensitive to crude oil exposure and thus, short term exposures during critical developmental periods could have detrimental consequences for juvenile survival. To obtain a broader understanding of the mechanisms behind crude oil toxicity, we administered crude oil to Atlantic haddock (Melanogrammus aeglefinus) in short term (3-day) exposures at two developmental time-points, before and after onset of heartbeat. A frequent sampling regime enabled us to determine and link immediate PAH uptake, metabolite formation and gene expression changes (cyp1a,b,c,d, bmp10, abcb1 and rhag). In general, the embryotoxic consequences were more severe in the earliest exposure, and oil droplet fouling enhanced the toxicity. This study detected circulation-independent, oil-induced gene expression changes and separated phenotypes linked to proliferation, growth and disruption of formation events at early and late developmental stages. A greater understanding of crude oil toxicity is essential for predicting the consequences of oil spills.Graphic abstractHighlightsOil droplet fouling occurred in the whole water column and increased the oil toxicity.Early exposure resulted in higher PAH uptake due to lower metabolism resulting in more severe abnormalities.A rapid and circulation-indepenent regulation of bmp10 suggested a direct oil-induced effect on calcium homeostasis.Expression of rhag indicated a direct oil-induced effect on osmoregulatory cells and osmoregulation.Severe eye abnormalities especially in the late exposure was linked to inappropriate overexpression of cyp1b in the eyes.

A new fast and effective analysis method has been developed to simultaneously determine 16 polycyclic aromatic hydrocarbons in reclaimed water samples by ultra-performance convergence chromatography with photodiode array detection and solid-phase extraction. The parameters of ultra-performance convergence chromatography on the separation behaviors and the crucial condition of solid-phase extraction were investigated systematically. Under optimal conditions, the 16 polycyclic aromatic hydrocarbons could be separated within 4 min. The limits of detection and quantification were in the range of 0.4-4 and 1-10 μg/L in water, respectively. This approach has been applied to a real industrial wastewater treatment plant successfully. The results showed that polycyclic aromatic hydrocarbons were dramatically decreased after chemical treatment procedure, and the oxidation procedure was effective to remove trace polycyclic aromatic hydrocarbons.
