Ondansetron is a serotonin receptor antagonist for treating nausea and vomiting. Its multiple daily doses and extensive first-pass metabolism faced parenteral and oral administration challenges. The transdermal route enhances its bioavailability as it bypasses the first-pass effect. Invasome vesicles improve the skin permeation of ondansetron via its unique components, lecithin, limonene, and ethanol, which are considered efficient permeation enhancers. The current study intends to develop a gel containing ondansetron as invasomes vesicles with appropriate texture and efficient skin penetration. Two gelling agents, hydroxypropyl methylcellulose K4M and carbopol 934P, were used at two concentrations (0.5 and 1%, w/w). Formulas were evaluated regarding pH, content uniformity, viscosity, spreadability, and ex vivo permeation. The selected formula (F3), which contained ondansetron (5%, w/w) and used carbopol 934P at (0.5%, w/w), was homogeneous, with proper viscosity (21,500 ± 390 mPa.s) at rest and enough spreadability (3.4 ± 0.45 cm). The ex vivo permeation showed permeation flux (Jss) was (280.4 ± 3.5 g/cm2.h) with a shorter lag time (0.5 ± 0.1 h). Characterization studies revealed nanosized vesicles (Dav. = 202.9 ± 0.5 nm) as spherical shapes dispersed within a gel matrix. In vivo, skin irritation and histopathology studies using male Wistar albino rats demonstrated that (F3) was biocompatible, with no irritation reported for six consecutive days. In conclusion, the invasomes gel of ondansetron utilizing carbopol (0.5%, w/w) was prepared for transdermal delivery, offering effective permeation and safe application. An invasomes-loaded gel is considered a novel formulation, giving an efficient and realistic therapeutic option for treating vomiting.

The purpose of this research work was to formulate & evaluate the herbal fast dissolving tablet of Nyctanthes Arbor Tristis. Taxonomically, the plant was authenticated by the Padmashri vikhe patil college of Arts, Science, & Commerce in pravaranagar. Ref.No./PVPC/Bot./2023-24/235. The aimed is to treat inflammation associated with Sciatica. A two independent variables of superdisintegrants are used such as crosspovidone & croscarmellose sodium. Using DOE software, 32 full factorial design was used to examine the combined impact of two formulation variables. TLC was performed to identify the beta sitosterol with RF value of 0.77. to determine the physicochemical interaction between the drug & the excipients, FTIR study was done. FTIR study revealed that there is no interaction of the drug and excipient. In this investigation, the tablets were formulated and assessed via direct compression. The powder mixture were compressed into tablet using single punch tablet machine. All the formulations were evaluated for their characteristic such as weight variation, hardness, thickness, friability, disintegration time(DT), and dissolution rate. It was determined from all evaluation parameter that the F9 batch was the optimized batch. It was found that an optimized batch (F9) has good cumulative drug release in 20 minutes (88.87%), DT of 56 seconds, and hardness of 2.923kg/cm2. The comparative studies are done with the marketed formulation. With the commercial formulation, the optimized batchF9 satisfies all standard specification.

Objective: The rationale of the current work is to design, develop and optimize of mouth dissolving tablet of ambrisentan to treat hypertension. Methods: Sodium starch glycolate and crospovidone were used as the super disintegrants in the direct compression method to create nine ambrisentan mouth-dissolving tablet formulations. Wetting time, drug content, in vitro disintegration time, dispersion time, and dissolution time were all assessed for the produced formulations. Results: Based on the results obtained, formulation F6 containing 30 mg of crospovidone exhibited good wetting time, dispersion time, disintegration time and drug release. The hardness of formulations AS1 to AS9 was found to be in the range of 2.5 to 3.11 Kg/cm2. The friability of formulations AS1 to AS9 was found to be less than 1%. A water absorption ratio was performed for ensuring the moisture sorption and water uptake properties of super disintegrants. The in vitro drug release of formulation AS6 containing a concentration of Crospovidone 30 mg, shows 91.30% drug release respectively at the end of 12 min. Conclusion: The mouth-dissolving tablets of ambrisentan were successfully designed, developed, and fabricated. It can be reasonably concluded that the AS6 batch of mouth-dissolving tablets of ambrisentan with 30 mg of crospovidone exhibited maximum cumulative drug release in 12 min.
