Four-directional stereo-microscopy for 3D particle tracking with real-time error evaluation

Abstract: Abstract:High-speed video stereo-microscopy relies on illumination from two distinct angles to create two views of a sample from different directions. The 3D trajectory of a microscopic object can then be reconstructed using parallax to combine 2D measurements of its position in each image. In this work, we evaluate the accuracy of 3D particle tracking using this technique, by extending the number of views from two to four directions. This allows us to record two independent sets of measurements of the 3D coor… Show more

“…Another promising approach used a spatial light modulator in reflection geometry to provide 3D information in an optical microscope ( [179], figure 17). Another simpler method uses two detectors that receive images from slightly different focal planes to synthesize 3D information (figure 18).…”

“…From a medical perspective, SM allows the visualization of microscopic surfaces magnified and dilated in all dimensions giving the observer an accurate sense of its real 3D topographical appearance (12,13,14) . Such perception allows observers to assess and precisely manipulate most specimens without the aid of specialized devices.…”

“…For example, the first necessary step for statistically analyzing the trajectory of a trapped particle is to track the particle position accurately. This is often achieved by acquiring a video 6,7 , stereomicroscopy 8 , V-shaped micro-mirrors 9 of the particle, or other visualization techniques including position sensitive detection 10 and quadrant photodiode detection 11 , which monitors via detecting the backscattering signal or transmitting signal. In fact, these experimental techniques are limited by sampling rate and in 3D imaging applications.…”
