Free Fatty Acids Regulate Two Galactosyltransferases in Chloroplast Envelope Membranes Isolated from Spinach Leaves

Abstract: Effects of MgCI2 and free fatty acids (FFA) on galactolipid:galactolipid galactosyltransferase (GGGT) and UDP-galactose:1,2-diacylglycerol galactosyltransferase (UDGT) in chloroplast envelope membranes isolated from spinach (Spinacia oleracea L.) leaves were examined. GGGT activity was sigmoidally stimulated by MgCI2 with a saturated concentration of more than 5 millimolar. Free a-linolenic acid (18:3) caused a drastic increase in GGGT activity under limiting concentrations of MgCI2, without affecting its maxi… Show more

“…Previous studies have demonstrated activation of TAG synthesis and a concurrent decrease in galactolipids and phospholipid in ozone-fumigated leaves (Sakaki et al, 1990a(Sakaki et al, , 1990b(Sakaki et al, , 1990c. Drought-stressed cotton leaves similarly exhibit a significant decline in polar lipid and a parallel increase in TAG (El-Hafid et al, 1989).…”

“…These authors postulated that GGGT provides the DAG precursor for triacylglycerol biosynthesis following ozone fumigation and produces oligogalactolipids in the process. Moreover, it was shown that GGGT was activated by free fatty acids in the presence of divalent cations such as Mg 2ϩ following ozone fumigation, whereas UDP-Gal-dependent galactosyltransferases were inhibited by free fatty acids under these conditions (51). Therefore one can speculate that GGGT plays a role during ozone-induced injury (Fig.…”

“…Transgenic plants were clearly under stress throughout growth and development as indicated by the consistent presence of anthocyanin in the leaves. Previous studies have demonstrated activation of TAG synthesis and a concurrent decrease in galactolipids in ozone-fumigated leaves (Sakaki et al, 1990a(Sakaki et al, , 1990b(Sakaki et al, , 1990c. Activation of TAG synthesis in the ozone-treated leaves is thought to reflect sequestration of fatty acids deesterified from galactolipids in response to ozone stress, which would otherwise disrupt the structure of thylakoid membranes (Sakaki et al, 1990b).…”
