Frequency of MDR1 single nucleotide polymorphisms in a Jordanian population, including a novel variant

Abstract: ABSTRACT. The multidrug resistance gene (MDR1 or ABCB1) codes for P-glycoprotein, which plays an important role in regulating absorption, distribution, and elimination of drugs. We examined MDR1 gene variants in 100 unrelated subjects from various regions of Jordan. The MDR1 gene was scanned using direct sequencing. Six rare variants in MDR1 were detected, including a new variant, T3075A. This variant did not affect the protein sequence (synonym for threonine). Among the common SNPs, the frequencies of rs11285… Show more

“…The distribution of G2677T/A and C1236T SNPs also shows variation among populations with high frequency of the mutant alleles among Africans and Asians (Cascorbi 2011;Gaikovitch et al 2003;Milojkovic et al 2011). We have recently reported that the MDR1 variants distribution among Jordanians is similar to that of Caucasians (Khabour et al 2012). …”

“…ABCB1 is involved in diseases and the efficacy of therapy, where SNPs in the ABCB1 gene affect its function. Polymorphisms of ABCB1 rs1128503 and rs1045642 are the most studied variants genetic of ABCB1 in diseases and indicate high frequencies in several populations [ 45 , 46 ]. This was the first genotype variation study of ABCB1 in Indonesian TB patients, especially in the Jambi population, although we did not find a significant association with sociodemography characteristics or duration of therapy.…”
