Freshly isolated hepatocyte transplantation in acetaminophen-induced hepatotoxicity model in rats

Abstract: -Context -Hepatocyte transplantation is an attractive therapeutic modality for liver disease as an alternative for orthotopic liver transplantation. Objective -The aim of the current study was to investigate the feasibility of freshly isolated rat hepatocyte transplantation in acetaminophen-induced hepatotoxicity model. Methods -Hepatocytes were isolated from male Wistar rats and transplanted 24 hours after acetaminophen administration in female recipients. Female rats received either 1x10 7 hepatocytes or pho… Show more

“…Hepatocytes were isolated from male Wistar rats with acute liver failure (n ¼6) or from healthy animals (n ¼6) by the two-step collagenase perfusion method as described by Seglen (Seglen, 1976), with modifications (Rodrigues et al, 2012), using collagenase type II (Gibco, USA). Hepatocytes were seeded in 6-well plates coated with type I collagen (3 Â 10 5 cells/well) and cultivated in low-glucose DMEM (Gibco) supplemented with 20% FBS (Gibco) and 1% P/S (Gibco) for 12 h to allow the cells to adhere before co-culture experiments.…”

“…For in vitro studies hepatocytes were isolated 24 hrs after CCl 4 using the modified 2-step perfusion method as previously described [30,31]. Hepatocytes from animals without liver injury were isolated by the same method.…”

“…Показано, что при трансплантации суспензии аллогенных изолированных гепатоцитов как в эксперименте [9], так и в клинике [10][11][12] удается достигнуть компенсации нарушенных функций печени. Однако степень выраженности возникающей компенсации и ее длительность определяются: с одной сторонытяжестью (обратимостью) исходного повреждения печени реципиента клеток, а с другойсроками сохранения жизнеспособности и регуляторной активности донорских гепатоцитов после их трансплантации.…”
