From Achiral Porphyrins to Template-Imprinted Chiral Aggregates and Further. Self-Replication of Chiral Memory from Scratch

Abstract: The presence of 10-13 M of chiral clusters of aromatic amino acids addresses aggregation of opposite-charged achiral porphyrin towards the formation of smart chiral assemblies. The latter supramolecular complexes are able to self-propagate and transfer their chiral information with a 100% yield. The chiral bias occurs through a correlated sequence of induction, memory, and amplification of chirality that strongly recalls possible prebiotic scenarios.

“…Taking advantage of the chiral memory approach, we have recently published the non-covalent synthesis of chiral porphyrin hetero-aggregates, templated by polypeptides or aminoacids. [16][17][18] These aggregates keep memory of the imprinted chirality that is maintained, even after removing the chiral template. Furthermore, their interactions in the absence of chiral templates leads to achiral species.…”

“…The structure of this complex originates from the geometry of polyglutamic acid in α-helical conformation, as is evident from the fact that the mirror image CD spectrum is obtained when poly-D-glutamate is used instead of poly-L-glutamate [9]. Porphyrin heteroaggregation has also been studied using phenylalanine as the chiral template [2,10,11]. This amino acid allows the formation of a chiral porphyrin heteroaggregate when it is used at a concentration in excess of 1 × 10…”

“…The most interesting phenomenon observed for porphyrin heteroaggregates is their self-replication ability [2,10,11]. This phenomenon is manifested as an ability of supramolecular systems to form structures with an identical arrangement when monomeric non-ordered units are added.…”

“…It has two negative charges and forms stable J-and H-aggregates depending on the pH and ionic strength in the system. The TPPS J-and H-aggregates shows the property of induced chirality in many systems, such as multilayer and Langmuir-Schaefer (L-S) films prepared from chiral and achiral molecules, [20][21][22][23] the solutions containing polylutamic acid, polylysine, cationic porphyrin etc., [24][25][26][27][28][29][30] and even by mechanical stirring in solutions. 31 However, there is no report about the reversible chirality of TPPS aggregates in film.…”
