Fructose-bisphosphate aldolase and pyruvate kinase, two novel immunogens inMadurella mycetomatis

Abstract: Eumycetoma, a chronic granulomatous disease characterized by a subcutaneous mass, multiple sinuses and purulent discharge containing grains, remains difficult to diagnose and treat. Madurella mycetomatis is the most common causative agent of eumycetoma. Using a serum pool from patients with active mycetoma, we screened a M. mycetomatis-specific λgt11 cDNA library which was shown to contain 8% of cDNA inserts encoding proteins involved in glycolysis. Two of these enzymes, fructose-bisphosphate aldolase (FBA) an… Show more

“…zooepidemicus through the blood-brain barrier. Finally, fructose-bisphosphate aldolase has been reported to be immunogenic in Candida albicans (Calcedo et al, 2012) and Madurella mycetomatis , in which it has been proposed as a vaccine candidate (de Klerk et al, 2012). …”

“…The only serological tools which use recombinant antigens are the ELISA described by van de Sande and colleagues [48] and the Luminex described by de Klerk and associates [49]. The recombinant translationally controlled tumour protein (TCTP) was included in both assays.…”

“…The recombinant translationally controlled tumour protein (TCTP) was included in both assays. The Luminex assay also included recombinant fructose-bisphosphate aldolase (FBA) and pyruvate kinase (PK) antigens [48], [49]. With these recombinant antigens it was noted that although patients gave higher antibody levels, healthy controls also produce antibodies against them, making it impractical to use these recombinant antigens as diagnostic tools.…”

“…Although the precise function of TCTP in M. mycetomatis has not yet been defined, several findings suggest it may play a role in mycetoma development. For one, the protein is only expressed in the very young grain and not in aged grains, unlike the other two antigens known for M. mycetomatis: fructose bisphosphate aldolase (FBA) and pyruvate kinase (PK) [28]. Such stage-specific expression may be associated with invasiveness.…”

“…This latter test employs recombinant TCTP or in vitro synthesized TCTP-specific peptides, derived from the first M. mycetomatis-specific antigen so far identified. Further novel M. mycetomatis-specific antigens are currently being identified and serological testing may be significantly improved in the near future [28].…”
