Functional analysis of −351 interleukin-9 promoter polymorphism reveals an activator controlled by NF-κB

Abstract: Genetic studies have shown linkages for asthma to the chromosomal region 5q31-q33 in humans that includes the IL-9 gene. An A-to-G base substitution has been identified at bp -351 in the IL-9 promoter. The role of this polymorphism in IL-9 promoter function was assessed utilizing CD4+ T cells purified from individuals with one or two of the G alleles in comparison to those homozygous for the wild type A. The presence of an A at -351 (A allele) increased mitogen-stimulated IL-9 transcription 2-fold in compariso… Show more

“…Each sub-type of T CD4+ affords protection, in general, to different types of infection [ 33 ]. NF-κB has been described as a crucial component in many of these sub-types of polarization (Th1, Th2, Th17, Th9) [ 34 , 35 , 36 , 37 , 38 , 39 ]. Box 1 summarizes the diverse effects of NF-κB on the immune system.…”

“…These results suggest that the noncanonical NF-κB pathway via the RelB:p52 heterodimer might be important in Th9 differentiation. Finally, it has been suggested that IL-9 expression in T cells is regulated by NF-κB, thereby implicating the pathway in the function of these cells (58). While many questions concerning the Th9 subset of CD4 + T cells remain unanswered, these findings represent the first steps towards a better understanding of the development and function of Th9 cells.…”

“…[7][8][9] Recent findings in the field have contributed to elucidate key molecular and cellular events involved not only in healthy immune response to allergens but also in breaking-tolerance to allergens or restoration of appropriate immune responses to allergens after AIT. [10][11][12][13][14][15][16][17] In this context, it is now well accepted that the generation and preservation of functional allergen-specific Treg cells are essential for healthy immune responses to allergens and successful AIT. [18][19][20] Treg cells comprise different subsets with suppressive capacity able to inhibit the initiation and development of allergic diseases and allergic patients seem to exhibit a specific impairment in the generation of regulatory T cells.…”
