Functional analysis of Pogostemon cablin farnesyl pyrophosphate synthase gene and its binding transcription factor PcWRKY44 in regulating biosynthesis of patchouli alcohol

Abstract: Farnesyl pyrophosphate synthase (FPPS) plays an important role in the synthesis of plant secondary metabolites, but its function and molecular regulation mechanism remain unclear in Pogostemon cablin. In this study, the full-length cDNA of the FPP synthase gene from P. cablin (PcFPPS) was cloned and characterized. The expressions of PcFPPS are different among different tissues (highly in P. cablin flowers). Subcellular localization analysis in protoplasts indicated that PcFPPS was located in the cytoplasm. PcF… Show more

“…Moreover, BBS belongs to sesquiterpene synthases that are localized in the cytosol. Our results are consistent with previous reports, such as A. thaliana [ 21 , 22 , 23 ], periwinkle [ 24 ], and Aquilegia species [ 25 ]. In vitro enzyme activity analysis suggested that MrFPS and MrBBS act as a functional FPS and α-bisabolol synthase, respectively.…”

“…34 Heterologous overexpression of transgenic PcFPPS in yeast cells and N. benthamiana significantly increased the content of terpenoids and promoted the biosynthesis of patchouli alcohol in synergy with WRKY44. 23 GpFPS1 and GpMYB81 synergistically promote the biosynthesis of gypenosides in Gynostemma pentaphyllum. 35 It can be seen that the FPS gene plays a key regulatory role in the secondary metabolism of various plants and promotes the accumulation of triterpenoids and the high yield of active ingredients.…”

“…16 To date, FPS has been cloned and characterized in a variety of plants, and it has been found that FPS1 and FPS2 genes play key roles during embryonic development in seeds of A. thaliana, and the LeFPS1 gene is enriched in young leaves of tomato, which is involved in the metabolism of farnesyl pyrophosphate, affecting cell division and enlargement. 17,18 In addition, the FPS genes in P. ginseng, 19 Artemisia annua, 20 Poria cocos, 21 Tripterygium wilfordii, 22 Pogostemon cablin, 23 Dendrobium nobile, 24 and B. platyphylla 25 were also cloned and verified.…”

“…Neophytadiene and phytol are diterpenoids, and little research has been done on their biosynthesis. In our previous study, OEL4 and OEL12 transgenic tobacco plants, which overexpressed PcFPPS, exhibited significantly higher levels of phytol and neophytadiene than wild-type lines (Wang et al, 2022). Based on the synthetic pathways involved in terpenoid biosynthesis, diterpenoid biosynthesis is widely thought to be related to MEP synthesis.…”
