Functional Characterization of CYP716 Family P450 Enzymes in Triterpenoid Biosynthesis in Tomato

Abstract: Triterpenoids are a group of structurally diverse specialized metabolites that frequently show useful bioactivities. These chemicals are biosynthesized from the common precursor 2,3-oxidosqualene in plants. The carbon skeletons produced by oxidosqualene cyclase (OSC) are usually modified by cytochrome P450 monooxygenases (P450s) and UDP-dependent glycosyltransferases. These biosynthetic enzymes contribute to the structural diversification of plant triterpenoids. Until now, many P450 enzymes have been character… Show more

“…This study contributes to an increased understanding of the role of CYP716A enzymes in the diversification of the triterpenoid backbone (Figure 4; Miettinen et al 2017;Misra et al 2017;Tamura et al 2017aTamura et al , 2017bYasumoto et al 2016Yasumoto et al , 2017. We also identified previously reported putative compounds, oleanolic, ursolic, and betulinic aldehydes (3, 7, 11), by synthesizing authentic standards and comparing their Rts and mass spectra with those of chromatographic peaks obtained from heterologous yeast extracts.…”

“…The CYP716A and CYP72A subfamilies are the primary CYP450 gene families involved in pentacyclic triterpenoid saponin diversification, with the CYP716A family being the largest multifunctional C28oxidase family involved in such oleanane-type triterpenoid saponins biosynthesis [16,27,28] (Fig. 8).…”

“…To further explore the functional roles and evolutionary relationships for these A. elata CYP450s, we next constructed a phylogenetic tree incorporating these 111 CYP540s along with 59 CYP450s from other plants, including A. thaliana (37), P. ginseng (20), Daucus carota (2) and Polygala tenuifolia (1).…”
