Functional Domains of the Nicotinic Acetylcholine Receptora

Abstract: The nicotinic acetylcholine receptor is a multisubunit, membrane-spanning protein that contains a gated, cation-conducting channel. Our approach to the understanding of the function of this receptor in molecular terms has been to locate its functionally significant sites in the sequences of its subunits and in its three-dimensional structure. In addition, we have tried to correlate transitions in the properties of these sites with functional transitions of the receptor. On binding acetylcholine, the nicotinic … Show more

“…This band was identified by immunoblotting as a modified form of the a subunit induced by irradiation (Fig. 2) 6, and 9), 20 ,uM a-BTX (lanes 4, 7, and 10), 30 ,uM histrionicotoxin (lanes 5-7), or 30 ,uM tetracaine (lanes 8-10). Proteins were separated by NaDodSO4/PAGE, stained with Coomassie blue (lane 1), and then processed for fluorography (lanes 2-10).…”

“…A 4.6-Å structure (1) shows five Ϸ160-Å-long rod-shaped subunits surrounding a central channel. From the extracellular side the subunits have a counterclockwise order of ␣␥␣␦␤ (2,3). Each subunit has a large extracellular Nterminal or ligand-binding domain followed by four transmembrane regions, M1-M4 (4).…”
