“…Interestingly, profiling of the spontaneous firing and membrane properties of ARC Drd1+ neurons revealed that, as a whole, they are less responsive than ARC AgRP/NPY+ neurons to fasting. Others have also measured the electrophysiological properties of ARC neurons given different experimental treatments, including fasting ( Liu et al., 2012 ), exercise ( Han et al., 2018 ), high fat diet (HFD) ( Baver et al., 2014 ; Wei et al., 2015 ; Jais et al., 2020 ), and gastric inputs ( Alhadeff et al., 2019 ; Goldstein et al., 2021 ; Jais et al., 2020 ). Ultimately, our results suggest the ARC Drd1+ neuronal population is heterogeneous.…”

“…The orexigenic effects of AgRP neurons are mediated by several AgRP neurocircuits that have been reviewed elsewhere (Deem et al, 2021; Han et al, 2018; Krashes, 2017; Sternson & Eiselt, 2017). Optogenetic stimulation of AgRP axonal projections to the BNST, LHA, MeA, PVH or the PVT drives food consumption (Figure 2) (Atasoy et al, 2012; Betley et al, 2013; Horio & Liberles, 2021; Padilla et al, 2016; Steculorum et al, 2016).…”

“…AgRP neurons are dynamically regulated across each feeding phase by signals that are internally released or externally encountered sequentially and/or that possess different temporal regulatory effects on AgRP neuronal activity. Besides promoting hunger and motivating an animal to seek food, AgRP neurons act as ‘gate‐keepers’ of survival by protecting mice from starvation through the initiation of a large spectrum of hunger‐related behavioural and physiological responses (reviewed in Deem et al, 2021; Han et al, 2018). For instance, AgRP neuron activation allows the animal to overcome environmental stressors and prioritise behaviours but also shuts down highly energy‐demanding processes.…”

“…Metabolic status and fertility are affected by many neuronal populations, one of which is the arcuate population of agouti‐related peptide (AgRP) neurons. These neurons drive food intake in response to situations of caloric need, and are modulated by metabolically‐relevant hormones such as leptin and insulin 1 . They also provide a link to the hypothalamic–pituitary‐gonadal (HPG) axis, possibly through functional synaptic contact with GnRH neurons 2–4 and/or with arcuate and anteroventral kisspeptin neurons 5,6 .…”
